摘 要:目的探讨肠道菌群紊乱及β-葡萄糖醛酸酶(β-Glucuronidase,GUS)与结直肠恶性肿瘤的相关性.方法收集肠癌、肠癌前病变患者及其同期健康人员粪便样本,进行细菌培养并计数.水煮法提取大肠埃希菌DNA后行PCR扩增,采用分子克隆技术构建重组质粒,进行DNA测序比对.结果细菌培养结果显示:大肠埃希菌、酵母菌、肠球菌、双歧杆菌、乳酸杆菌在肠癌、肠癌前病变患者及健康对照3组间比较差异具有统计学意义(P<0.05).测序结果显示:肠癌前病变患者大肠埃希菌uidA基因序列第1497位置G突变成A.结论肠癌前病变患者肠道菌群失调在癌变早期已发生改变,大肠埃希菌uidA基因存在突变,肠癌的发生风险增加.Objective The disorder of intestinal flora and the activity alteration of beta-glucuronidase(GUS)was closely related to colorectal cancer.The aim of this study was to provide new idea for early diagnostic indexes of colorectal cancer and to predict the risk of carcinogenesis.Method Collecting stool samples from the patients of colorectal cancer and precancerous lesions and healthy persons at the same time to culture and count intestinal bacterias.PCR amplification after extraction of Escherichia coli DNA by water-boiling method,the recombinant plasmid was constructed by molecular cloning technique then compared by DNA sequencing.Results The results of bacterial culture showed that the difference of Escherichia coli,yeast,Enterococcus,Bifidobacteria,Lactobacillus in the patients of colorectal cancer and precancerous lesions and healthy controls was statistically significant(P < 0. 05). The results of sequencing showed uidA gene from Escherichia coli in patients with precancerous lesions G mutated to A in position 1497. Conclusion In the early stage of carcinogenesis,the alteration of intestinal flora had occurred in patients with precancerous lesions,uidA gene from Escherichia coli existed mutation and the risk of colorectal cancer was increased.
