摘 要:目的:研究姜黄素对增殖性玻璃体视网膜病变(PVR)大鼠模型的治疗效果及对SDF-1/CXCR4轴的影响。方法:建立PVR大鼠模型,并用裂隙灯、间接检眼镜进行验证,按照实验分组处理各组大鼠,眼底彩色照相系统、B型超声和视网膜HE染色评价处理后各组大鼠PVR缓解情况,蛋白印迹法(WB)测定各组大鼠视网膜组织SDF-1、CXCR4蛋白表达水平,酶联免疫法(ELISA)法检测各组大鼠血清中IL-6、IL-1β、IL-8、TNF-α表达水平。结果:裂隙灯、间接检眼镜证明高PVR大鼠模型构建成功,与对照组相比,PVR模型组大鼠视网膜玻璃体浑浊,可见增生条带,可见视网膜漏斗状或全脱离,内外核层、神经节细胞排列混乱,细胞数量减少,结构破坏,视网膜脱离眼数显著升高(P<0.05),SDF-1、CXCR4、IL-6、IL-1β、IL-8、TNF-α表达水平显著升高(P<0.05);与高氧模型组相比,低、中、高剂量姜黄素组、妥布霉素地塞米松滴眼液组视网膜病变程度逐渐缓解,玻璃体浑浊程度降低,内外核层、神经节细胞排列分明,结构恢复正常,视网膜脱离眼数逐渐显著下降(P<0.05),SDF-1、CXCR4、IL-6、IL-1β、IL-8、TNF-α表达水平显著降低(P<0.05),且呈剂量依赖性,高剂量组PVR缓解程度与妥布霉素地塞米松滴眼液组相当。结论:姜黄素可能通过抑制SDF-1/CXCR4轴活化,缓解PVR大鼠模型症状。Objective:To study the therapeutic effect of Curcumin on proliferative vitreoretinopathy(PVR) rat model and its influence on SDF-1/CXCR4 axis.Methods:The rat model of PVR was established and validated by slit lamp and indirect ophthalmoscope,the rats in each group were treated according to the experimental group,the relief of PVR was evaluated by fundus color photography system,B-mode ultrasound and HE staining,the expression levels of SDF-1 and CXCR4 proteins in retina of rats in each group were determined by Western Blotting(WB),and the expression levels of IL-6,IL-1β,IL-8 and TNF-α in serum of rats in each group were detected by enzyme-linked immunosorbent assay(ELISA).Results:Slit lamp and indirect ophthalmoscope proved that the rat model with high PVR was successfully constructed,compared with the controlled group,retinal vitreous opacity occurred in rats in PVR model group,proliferation bands,funnel-shaped or complete detachment of the retina could be seen,the arrangement of inner and outer nuclear layer and ganglion cells was disordered,the number of cells decreased,the structure was destroyed,and the number of eyes with retinal detachment increased significantly(P < 0.05),the expression levels of SDF-1,CXCR4,IL-6,IL-1β,IL-8 and TNF-α increased significantly(P < 0.05);Compared with the hyperoxia model group,the degree of retinopathy in the low,medium and high dose Curcumin group and Tobramycin and Dexamethasone Eye Drops group gradually alleviated,the vitreous opacity decreased,the inner and outer nuclear layers and ganglion cells arranged clearly,the structure returned to normal,and the number of eyes with retinal detachment decreased significantly(P < 0.05),the expression levels of SDF-1,CXCR4,IL-6,IL-1β,IL-8 and TNF-α decreased significantly(P < 0.05),which were dose-dependent,the alleviation of PVR in the high dose group was similar to that in the Tobramycin and Dexamethasone Eye Drops group.Conclusions:Curcumin may alleviate the symptoms of PVR rat model by inhibiting the activation of SDF-1/CXC
