蛋氨酸缺乏对雏鸡肝细胞周期的影响

摘　　要：为了研究日粮蛋氨酸缺乏对雏鸡肝脏的细胞周期、细胞周期调控蛋白及基因的影响,本研究选用1日龄雌性雏鸡72只,随机分为对照组和蛋氨酸缺乏组,实验期为42 d。利用流式细胞术检测肝细胞周期变化、酶联免疫吸附法(ELISA)测定细胞周期调控蛋白(p53、p21、CyclinB、PCNA)的含量以及荧光定量聚合酶链式反应法(RTFQ-PCR)分析其基因表达变化。结果显示,相较于对照组,蛋氨酸缺乏组肝细胞G0G1期和S期百分比显著下降,而G2M期显著或极显著上升;p53和p21蛋白量显著或极显著上升趋势,CyclinB蛋白和PCNA蛋白量却显著或极显著下降;周期相关基因表达结果与蛋白一致。由此得出结论:蛋氨酸缺乏会导致肝细胞G2M期阻滞,使其增殖受抑制。The purpose of this study is to investigate the effects of methionine(Met)deficiency on hepatic cell cycle,cell cycle regulatory proteins and cyclin-related genes of broiler chicken.All of 721-day-old female broilers are randomly divided into two groups and fed on a Met deficiency diet or a control diet for 42d.The percentage of cells in each phase is determined by flow cytometer(FCM),and the content of cell cycle regulatory proteins(P53,P21,CyclinB,PCNA)are determined by Enzyme-linked Immunosorbent Assay(ELISA).The expression of genes are analyzed by realtime fluorescence quantitative PCR(RTFQ PCR).The results indicate that when compared with the control group,G0G1 and S phases of hepatic cell have significantly decreased but G2M has increased or notably increased;P53 and P21 have shown a marked or extremely marked upward trend than that of control group but CyclinB protein and PCNA protein levels have decreased significantly or extremely.The expression of cycle-related genes are consistent with that of corresponding proteins.In conclusion,methionine deficiency can induce G2M phase arrest and inhibit proliferation of broiler chicken.
