摘 要:以多肉植物康平寿(Haworthia emelyae)为试材,采用植物组织培养法,Cd2+浓度为0.01、0.05、0.10、0.50和1.00 mmol/L,研究Cd2+对康平寿试管苗生长和增殖的影响,为筛选有效的Cd污染修复植物提供参考依据.结果表明:经0.01~0.05 mmol/L Cd2+处理后,试管苗的植株高度、鲜重和增殖系数与对照相比,均无显著差异(P>0.05);经0.10~0.50 mmol/L Cd2+处理后,试管苗的植株高度、鲜重和增殖系数显著低于对照(P<0.05);当Cd2+浓度为0.50 mmol/L时,试管苗的增殖系数为1.00,其增殖能力被完全抑制;当Cd2+浓度达0.05 mmol/L时,试管苗产生的不定根的数量和长度显著低于对照(P<0.05);试管苗在1.00 mmol/L Cd2+浓度作用下,第3周开始发生枯黄,最后全部死亡.分析表明:多肉植物康平寿对重金属镉具有一定的耐受性,低浓度的Cd对康平寿的生长发育无显著影响,但高浓度的Cd对植株的生长发育具有抑制作用,并且其抑制作用随着Cd浓度的升高愈发显著.Taking Haworthia emelyae as materials,the effects of Cd2+at different concentrations of 0.01,0.05,0.10,0.50 and 1.00 mmol/L on the growth and proliferation of Haworthia emelyae shoots in vitro were studied through plant tissue culture,so as to provide certain basis for screening effective Cd-restoring plants.The results showed that there were no significant changes in plant height,fresh weight and proliferation coefficient of the shoots treated with 0.01~0.05 mmol/L Cd2+when compared with the control.After treatment of 0.10~0.50 mmol/L Cd2+,the plant height,fresh weight and proliferation coefficient of the shoots significantly decreased(P<0.05);the proliferation coefficient of shoots when treated with 0.5 mmol/L Cd2+was 1.0,indicating that the proliferation ability was completely inhibited.The number and length of adventitious roots from shoots decreased significantly(P<0.05)after treatment with≥0.05 mmol/L Cd2+.The shoots started turning yellow after two weeks of treatment with 1.00 mmol/L Cd2+,and all died after 40 days.The results indicate that Haworthia emelyae had certain tolerance on heavy metal cadmium,lower concentration of Cd had no significant impact on the growth and development of Haworthia emelyae,higher concentration of Cd significantly inhibited the growth and development,and the inhibitory effect increased with the concentration of cadmium.
