摘  要:为证实室旁核(PVN)自噬介导慢性心衰(CHF)大鼠交感神经兴奋性亢进,冠动脉结扎诱导CHF,在PVN内慢性灌注自噬促进剂RAPA或抑制剂3-MA,观察大鼠心脏功能及交感神经驱动指标的变化,并检测PVN内LC3-Ⅱ蛋白表达量.结果表明,与CHF溶剂组相比,灌注RAPA导致CHF大鼠交感驱动指标升高,心功能下降;相反,灌注3-MA导致CHF大鼠的交感驱动指标下降,心功能得到改善;与假手术(Sham)相比,CHF组PVN中LC3-Ⅱ蛋白的表达显著上调.上述结果表明PVN自噬水平增强介导了CHF大鼠交感神经兴奋性亢进,可进一步恶化CHF大鼠的心功能.In order to confirm that paraventricular nucleus(PVN)autophagy mediates sympathetic overactivity in rats with chronic heart failure(CHF),the animal model of CHF was induced by coronary artery ligation.The chronic perfusion of autophagy agonist RAPA or inhibitor 3-MA in PVN was used to observe the changes of cardiac function and sympathetic drive index in rats,and the LC3-Ⅱprotein expression in PVN was detected.The results showed that compared with CHF vehicle group,perfusion of RAPA resulted in increased sympathetic drive index and decreased cardiac function in rats with CHF.On the contrary,perfusion of 3-MA resulted in decreased sympathetic drive index and improved cardiac function in rats with CHF.Compared with sham operation,the expression of LC3-Ⅱ protein in PVN of CHF rats was significantly upregulated.The above results suggest that the enhancement of PVN autophagy level mediates sympathetic overactivity,and may further deteriorate cardiac function in rats with CHF.
