摘 要:目的探讨苏格木勒-3汤治疗心力衰竭的可能作用机制。方法 HL-1心肌细胞按每孔1×10~5个细胞接种于96孔板,常规培养24h后进行分组:空白对照组的不加任何处理的正常心肌细胞;模型组加入终浓度为25μg/ml的异丙肾上腺素每孔100μl处理24h;苏格木勒-3汤高、中、低剂量组加入终浓度为25μg/ml异丙肾上腺素每孔100μl处理24h后分别加入75、50、25μg/ml苏格木勒-3汤水提液每孔100μl处理24h;苏格木勒-3汤对照组加入75μg/ml苏格木勒-3汤水提液每孔100μl处理24h。每组5个复孔。MTT法检测各组HL-1心肌细胞存活率,荧光显微镜检测细胞中钙离子浓度,Western blotting法检测HL-1心肌细胞中肌浆网钙ATP酶2a (SERCA2a)蛋白表达。结果与空白对照组比较,模型组HL-1心肌细胞存活率、SERCA2a蛋白表达下降(P<0.01)。与模型组比较,苏格木勒-3汤高、中剂量组HL-1心肌细胞存活率增高,苏格木勒-3汤高、中、低剂量组HL-1心肌细胞SERCA2a蛋白表达量均增加(P<0.01)。荧光显微镜下显示,模型组HL-1心肌细胞内钙离子浓度的荧光强度增加;与模型组比较,苏格木勒-3汤中、高剂量组对HL-1心肌细胞内钙离子浓度的荧光强度逐渐下降。结论蒙药苏格木勒-3汤对HL-1心肌细胞钙超载具有抑制作用,其机制可能与提高心肌细胞中SERCA2a蛋白表达有关。Objective To explore the possible mechanism of Sugemule-3 Decoction(苏格木勒-3汤) in the treatment of heart failure. Methods HL-1 cardiomyocytes were inoculated into 96-well plates at 1×10~5 cells per well, and they were grouped after 24 h of routine culture. In blank control group, there were normal cardiomyocytes without any treatment. Model group was added with 100μl isoproterenol per well with a final concentration of 25μg/ml to process for 24 h. After being added with 100μl isoproterenol per well with a final concentration of 25μg/ml to process for 24 h, Sugemule-3 Decoction groups of high, medium and low doses were added with 75, 50, and 25μg/ml water extract of Sugemule-3 Decoction, respectively, to process for 24 h, with 100μl per well. There were 5 double wells in each group. MTT method was used to detect the survival rate of HL-1 cardiomyocytes in each group. Calcium ion concentration was detected by fluorescence microscope. Western blotting method was used to detect protein expression of sarcoplasmic reticulum calcium ATPase 2 a(SERCA2 a) in HL-1 cardiomyocytes. Results Compared with the blank control group, the survival rate of HL-1 cardiomyocytes and the expression of SERCA2 a protein in the model group were decreased(P<0.01). Compared with the model group, the survival rates of HL-1 cardiomyocytes in the high and medium dose groups of Sugemule-3 Decoction were increased, and the expressions of SERCA2 a protein in HL-1 cardiomyocytes of the high, medium and low dose groups of Sugemule-3 Decoction were increased(P<0.01). Fluorescence microscope showed that the fluorescence intensity of calcium ion concentration in HL-1 cardiomyocytes of the model group was increased;compared with the model group, the fluorescence intensities of calcium ion concentration in HL-1 cardiomyocytes in the medium and high dose groups of Sugemule-3 Decoction were decreased.
