摘  要:目的:优化瑶药地耳草总黄酮的提取工艺,并建立其药材的提取和含量测定方法。方法:采用回流提取法提取地耳草总黄酮,通过单因素试验,考察提取方法、提取溶剂浓度、提取体积、提取时间对其药材总黄酮含量的影响,采用正交试验,优化提取工艺条件,筛选出地耳草总黄酮的最佳提取工艺。结果:地耳草总黄酮的最佳提取工艺为70%乙醇、料液比1:40、回流提取时间1.0h。结论:该方法能有效测定地耳草药材总黄酮的含量,为该药材的质量标准研究提供一定的科学依据。本试验提取地耳草药材总黄酮含量,重复性较好、方法稳定、可行。Objective:To optimize the extraction process of total flavonoids from Hypericum japonicum Thumb.,and to establish a method for the extraction and content determination of the total flavonoids.Methods:The total flavonoids of Hypericum japonicum Thumb.were extracted by reflux extraction method.The effects of extraction method,extraction solvent concentration,extraction volume and extraction time on the total flavonoids content of the medicinal materials were investigated by single factor experiment.The orthogonal test was used to optimize the extraction conditions.The best extraction process for a total flavonoid of Hypericum japonicum Thumb.was screened out.Results:The optimal extraction process of a total flavonoid of Hypericum japonicum Thumb.was 70%methanol,the ratio of material to liquid was 1:40,and the reflux extraction time was 1.0h.Conclusion:This method can effectively determine the content of total flavonoids in Hypericum japonicum Thumb.medicinal material,and provide a scientific basis for the quality standard research of this medicinal material.In this experiment,the total flavonoid content of Hypericum japonicum Thumb.medicinal material was extracted,the repeatability was good,the method was stable and feasible.
