摘 要:目的探讨白藜芦醇(RES)能否通过抗氧化应激起到减轻小鼠急性肺损伤(ALI)的作用。方法采用随机数字表法将30只小鼠分为对照组(C组)、ALI组和RES干预组(RES组)3组,每组10只。RES组给予RES(50mg/kg体重)灌胃,1次/d,连续3d。第4天时,ALI组和RES组腹腔注射脂多糖(LPS)10mg/kg制备内毒素诱发ALI模型,C组则腹腔内注射同等容量的0.9%氯化钠注射液。6h后麻醉开腹,从下腔静脉取静脉血,采用ELISA法检测血清中肿瘤坏死因子α(TNF-α)和白细胞介素-6(IL-6)水平。光镜下观察肺组织病理学结果,并进行肺损伤评分。采用比色法检测肺组织匀浆中髓过氧化物酶(MPO)、超氧化物歧化酶(SOD)含量。结果与C组比较,ALI组和RES组肺组织病理学损伤评分、血清TNF-α、IL-6水平及肺组织MPO含量均升高,肺组织SOD含量均降低,差异均有统计学意义(均P<0.05)。与ALI组比较,RES组肺组织病理学损伤评分、血清TNF-α、IL-6水平及肺组织MPO含量均降低,肺组织SOD含量升高,差异均有统计学意义(均P<0.05)。结论RES可使LPS诱导ALI模型小鼠血清促炎症介质水平和组织中氧化应激损伤指标(MPO)降低,从而减轻ALI的严重程度。Objective To investigate the effect of resveratrol(RES)on acute lung injury(ALI)in mice and its mechanism.Methods Thirty healthy mice were divided into three groups:control group(group C),ALI group and the RES group.RES group was given RES orally q.d for 3d.On the D4,ALI group and the RES group were intraperitoneally injected with lipopolysaccharide(LPS)to induce acute lung injury,while group C was given same volume of normal saline instead.The levels of IL-6 and TNF-α in serum was measured by ELISA 6h after the injection of LPS or normal saline.The mice were then sacrificed,and the pathological changes in lung tissue were observed under light microscope to get the lung injury scores.The activity of MPO and SOD in lung tissue was measured by colorimetric method.Results Compared with the group C,the lung injury score and the levels of IL-6,TNF-α,MPO was increased,while the level of SOD was decreased significantly in ALI group(all P<0.05).Compared with the ALI group,the lung injury score and the levels of IL-6,TNF-α,MPO was reduced,while the level of SOD was increased significantly in RES group(all P<0.05).Conclusion Resveratrol can attenuate the severity of acute lung injury induced by LPS,reduce the concentration of proinflammatory mediators in serumand the oxidative stress index in the tissues.
