用高效液相色谱法测定胃乐胶囊中木香烃内酯和去氢木香内酯含量的效果分析

摘　　要：目的:研究用高效液相色谱(HPLC)法测定胃乐胶囊中木香烃内酯和去氢木香内酯含量的效果。方法:采用HPLC法测定胃乐胶囊中木香烃内酯和去氢木香内酯的含量,进行测定时的色谱条件是:色谱柱为GL Inertsil ODS-3,流动相为甲醇-水(65:35),流速为1.0 ml/min,检测波长为225 nm,柱温为25℃。在采用该法完成测定后观察测定的结果。结果:用HPLC法对三批样品进行测定的结果显示,胃乐胶囊中木香烃内酯的平均含量为0.048mg/g,去氢木香内酯的平均含量为0.059 mg/g。木香烃内酯和去氢木香内酯的进样量在0.200~4.002 μg的范围内与峰面积呈良好的线性关系。对样本进行精密度试验的结果显示,木香烃内酯和去氢木香内酯峰面积的RSD分别为0.91% 和0.96%。对多批样本进行重复性试验的结果显示,木香烃内酯和去氢木香内酯峰面积的RSD分别为1.75%和1.63%。对供试品溶液进行稳定性试验的结果显示,木香烃内酯和去氢木香内酯峰面积的RSD分别为0.64%和0.57%。对样本进行加样回收率试验的结果显示,木香烃内酯的平均加样回收率为99.7%,去氢木香内酯的平均加样回收率为102.2%,二者的RSD分别为1.7%和1.5%。结论:用HPLC法测定胃乐胶囊中木香烃内酯和去氢木香内酯的含量具有精密度、重复性和稳定性良好等优点。objective:to study the effect of high performance liquid chromatography(HPLC)for the determination of xylolactone and dehydroxylactone in weilei capsule.Methods:HPLC method was used to determine the contents of ligninolactone and dehydrocostolactone in weilei capsules.The chromatographic conditions were as follows:the chromatographic column was glinertsil ods-3,the mobile phase was methanol-water(65:35),the flow rate was 1.0 ml/min,the detection wavelength was 225 nm,and the column temperature was 25℃.The results were observed after the method was used.Results:the results of three batches of samples by HPLC showed that the average content of xylolactone in weilei capsule was 0.048mg/g,and that of dehydroxylactone was 0.059 mg/g.There was a good linear relationship between the injection amount and the peak area in the range of 0.200~4.002 g.The results of precision test on the samples showed that the RSD of the peak area of costolactone and dehydrocostolactone were 0.91%and 0.96%respectively.The results of repeated tests on multiple batches of samples showed that the RSD of the peak area of costolactone and dehydrocostolactone were 1.75%and 1.63%respectively.The results of the stability test of the sample solution showed that the RSD of the peak area of xylonolactone and dehydroxylonolactone were 0.64%and 0.57%,respectively.The results of recovery test showed that the average recovery was 99.7%and 102.2%respectively,respectively.Conclusion:HPLC method for the determination of xylolactone and dehydroxylactone in weile capsule has the advantages of precision,repeatability and good stability.
