摘 要:目的分别通过胰蛋白酶、糜蛋白酶、透明质酸酶、菠萝蛋白酶对精液进行处理,观察液化情况,记录液化时间,染色后镜下观察对精子形态的影响。方法每例精液标本均分别设立为五组,即对照组(不加处理因素)、胰蛋白酶组、糜蛋白酶组、透明质酸酶组、菠萝蛋白酶组,观察精液液化情况、并记录液化时间。筛选出液化正常精液标本30例,液化异常精液30例,待标本完全液化后,瑞氏染色,选择两名有经验的工作人员双盲法按照第五版《世界卫生组织人类精液检查与处理实验室手册》所规定的方法,分别记录头部、颈部和中段、主段的畸形率,对精子形态进行分析。结果液化异常精液组中加入四种酶后,各酶处理组精液液化时间较对照组显著缩短,差异有统计学意义(P<0.01)。正常液化精液组:加入胰蛋白酶后,头部畸形率明显升高,出现大头、小头、锥形头、梨形头等;加入透明质酸酶后精子颈部和中段、主段畸形率明显升高,颈部和中段出现粗或不规则,主段出现断裂,成角弯曲,卷曲等,主段、颈部和中段,差异均有统计学意义(P<0.01)。液化异常精液组:加入胰蛋白酶后,主段出现断裂,成角弯曲,卷曲等,差异有统计学意义(P<0.01);加入透明质酸酶后颈部和中段畸形率、主段畸形率明显升高,颈部和中段出现粗或不规则,主段出现断裂,成角弯曲,卷曲等,主段、颈部和中段,差异均有统计学意义(P<0.01)。其余各酶处理组各部畸形率与对照组比较差异均无统计学意义(P>0.05)。结论通过加入糜蛋白酶或菠萝蛋白酶来缩短精液液化时间,改善精液液化情况,进一步提高液化异常精液精子形态学分析的可靠性。Objective The liquefied semen was treated with trypsin,chymotrypsin,hyaluronidase and bromelain,and the liquefaction of semen was observed and the time of liquefaction was recorded.The effects of film and staining on the morphology of sperm were observed.Methods The sample was set up in five groups,that is,the control group(without any treatment),trypsin group,chymotrypsin group,hyaluronidase group,and bromelain.Group.Observation of semen liquefaction and recording of semen liquefaction time.30 specimens of Abnormal Liquefaction and 30 normal specimens of liquefaction were collected.After the specimen was completely liquefied,smear and filmmaking were stained with Rayleigh staining,and two experienced staff were selected under double blind microscope and the morphology of sperm was analyzed.Results The liquefying time:after adding four enzymes in the normal semen group,there was no significant difference in the time of semen liquefaction between the control group and the control group(P>0.05).After adding four enzymes in the group of liquefied abnormal semen,compared with the control group,the liquefying time of the semen of each enzyme treatment group was less than 30min and significantly shortened,the difference was statistically significant(P<0.01).After adding trypsin in the normal liquified semen group,the sperm appeared large head,small head,conical head,pear shaped head and other head deformities,and the rate of sperm head deformity increased obviously,the difference was statistically significant(P<0.01).After adding hyaluronidase,the sperm neck and middle segment,the main segment deformity rate increased obviously,and the neck and middle segment appeared rough or irregular.The main segment was broken,angled,curled,and the difference was statistically significant in the main segment,neck and middle part(P<0.01).However,chymotrypsin and bromelain had no significant effect on sperm morphology in normal liquefied semen,and the difference was not statistically significant(P>0.05).After adding trypsin in the ab
