摘 要:目的研究亚硒酸钠对醋酸铅致TK6细胞氧化应激损伤的保护效应。方法培养TK6细胞,调整细胞密度为1×106/ml,试验分3组,正常对照组、醋酸铅染毒组和亚硒酸钠干预组,采用流式细胞仪检测细胞内活性氧簇(ROS)水平,硫代巴比妥酸(TBA)法检测细胞内丙二醛(MDA)含量和高度水溶性四唑盐(WST-1)法检测细胞内超氧化物歧化酶(SOD)活性。结果在醋酸铅染毒和0.01μg/ml的亚硒酸钠干预后,3组细胞内ROS含量、MDA含量和SOD含量的比较,差异均有统计学意义(F分别为36.706,0.050,23.051,均P<0. 01),其中醋酸铅染毒后,TK6细胞内ROS含量和MDA含量增高,与正常对照组比较差异有统计学意义(P<0.01),0.01μg/ml的亚硒酸钠干预后,细胞内ROS含量和MDA含量下降,与醋酸铅染毒组比较差异有统计学意义(P<0.01);醋酸铅染毒后,TK6细胞内SOD含量降低,与正常对照组比较差异有统计学意义(P<0.01),0.01μg/ml的亚硒酸钠干预后,SOD含量上升,与正常对照组比较差异无统计学意义(P>0.05),而与醋酸铅染毒组比较差异有统计学意义(P<0.01)。结论醋酸铅染毒对TK6细胞有氧化损伤作用,0.01μg/ml亚硒酸钠对醋酸铅氧化损伤毒性具有拮抗作用。Objective To study the protective effect of sodium selenite on the oxidative damage to TK6 cells induced by lead.Methods TK6 cells were cultured and the cell density was adjusted to 1×106/ml.The cells were divided into 3 groups:normal control group,lead exposed group and sodium selenite intervention group.Then the intracellular reactive oxygen species(ROS)levels were detected by flow cytometry.Meanwhile,the content of malondialdehyde(MDA)was detected by thiobarbituric acid(TBA)assay and the activity of total superoxide dismutase(SOD)was detected by WST-1 method.Results After the intervention of lead acetate and 0.01μg/ml sodium selenite,there were statistically significant differences in ROS content,MDA content and SOD content in three groups of cells(respectively,F=36.706,F=40.050,F=23.051,all P<0.01).After lead exposure,the level of ROS and the content of MDA in TK6 cells were higher than that in the normal control group(P<0.01).The intracellular ROS level and MDA content decreased after treatment with sodium selenite at 0.01μg/ml,and compared with lead exposure group,the difference was statistically significant(P<0.01).The activity of SOD in TK6 cells decreased after lead exposure,and the difference was statistically significant compared with the control group(P<0.01),but the activity of SOD increased after treatment with sodium selenite at 0.01μg/ml,which was significantly different from lead exposure group(P<0.01),and had no significant difference from that of normal control group(P>0.05).Conclusion Lead has oxidative damage effect on TK6 cells,and 0.01μg/ml sodium selenite can protect TK6 cells from oxidative damage induced by lead.
