调心安神针法预处理对缺血再灌注性心律失常兔心肌细胞凋亡的影响

摘　　要：目的:研究调心安神针法预处理对于缺血再灌注性心律失常(RA)兔心肌细胞凋亡的影响。方法:采用结扎/松解兔左冠状动脉前降支的方法制作RA模型兔。40只健康家兔随机分为假手术组、模型组、针刺组、胺碘酮组,针刺组施以调心安神针刺法(取穴:灵台、神道、双侧内关、百会)。实验完毕后取兔缺血区中央部位心内膜心肌,免疫组织化学染色技术及计算机图像处理系统分析Bcl-2、Bax的表达水平,Western印迹法检测活性Caspase-3的表达水平。结果:针刺组、胺碘酮组Bcl-2的表达均较模型组明显升高(P<0.01),Bax、活性Caspase-3的表达均较模型组降低(P<0.01),胺碘酮组Bcl-2、活性Caspase-3的表达较针刺组变化更为显著(P<0.05或P<0.01)。结论:调心安神针法预处理对RA模型兔心肌细胞的凋亡有抑制作用。Objective:To observe the effect of Tiaoxin Anshen acupuncture on apoptosis in myocardial cells of ischemic reperfusion arrhythmia rabbits.Methods:40 rabbits were randomly divided into the sham operation group,the model group,the acupuncture group and the Amiodarone group.The acupuncture group was treated with Tiaoxin Anshen acupuncture(Needling GV10,GV11,bilateral PC6,GV20).After completion of the experiment,the central part of the endocardium of the ischemic area was taken out to detect the expressions of Bcl-2 and Bax by immunohistochemical technique and computer image processing system,and western blot method was used to detect the expression level of Caspase-3.Results:The expressions of Bcl-2 in the acupuncture group and in the Amiodarone group were significantly higher than those in the model group(P<0.01),and the expressions of Bax and Caspase-3 were significantly lower in the two groups than those in the model group(P<0.01);the expressions of Bcl-2 and Caspase-3 in the Amiodarone group were more significant than those in the acupuncture group(P<0.05,P<0.01).Conclusion:The pretreatment of Tiaoxin Anshen acupuncture has inhibiting effect on the apoptosis of rat myocardial cells in RA model rabbits.
