摘 要:目的观察补阳还五汤对Cav-1^-/-小鼠脑缺血模型PI3K、Akt、PTEN表达的影响,探讨其可能的作用机制。方法将野生型小鼠(WT)及Cav-1^-/-小鼠(KO)随机分为假手术组、模型组与补阳组。采用大脑中动脉栓塞法制备脑缺血模型,干预10 d后运用Ayelet Levy 14分评分法观察小鼠神经功能评分。免疫组化和qRT-PCR分别检测小鼠脑组织PI3K、Akt、PTEN蛋白和mRNA的表达。结果与假手术组比较,各模型组小鼠均出现明显神经功能障碍(P<0.01),且PI3K、Akt蛋白和mRNA表达明显升高(P<0.01),PTEN蛋白和mRNA表达明显降低(P<0.01),WT模型组小鼠脑组织PI3K、Akt、PTEN蛋白和mRNA表达更显著,且神经功能评分优于KO模型组(P<0.01,P<0.05);WT补阳组小鼠脑组织PI3K、Akt、PTEN蛋白和mRNA的调控程度及神经功能恢复程度均优于WT模型组和KO补阳组(P<0.01)。结论补阳还五汤可能通过Cav-1调控PI3K/Akt信号通路活性,发挥抗脑缺血损伤的作用。Objective To observe the effects of Buyang Huanwu Decoction on expressions of PI3K,Akt and PTEN after cerebral ischemia in Cav-1^-/-mice;To discuss its possible mechanism.Methods Wild type mice(WT)and Cav-1^-/-mice(KO)were randomly divided into sham-operation group,model group and Buyang Huanwu Decoction group.Cerebral ischemia model was established by middle cerebral artery occlusion.After 10 days of intervention,the neurological function scores of each group were observed by Ayelet Levy 14 points.Immunohistochemistry and qRT-PCR were used to detect the expressions of PI3K,Akt,PTEN protein and mRNA.Results Compared with sham-operation group,there were obvious neurological dysfunction in each model group(P<0.01),and the expressions of PI3K,Akt protein and mRNA increased significantly(P<0.01),PTEN protein and mRNA decreased significantly(P<0.01).Besides,PI3K,Akt,PTEN proteins and mRNA expression in WT model group was more significant and the neurological function score was better than that of KO model group(P<0.01,P<0.05).In addition,the degree of protein and mRNA regulation and neurological function recovery in WT Buyang Huanwu Decoction group was better than that in WT model group and KO Buyang Huanwu Decoction group(P<0.01).Conclusion Buyang Huanwu Decoction may play an anti-cerebral ischemic injury role by regulating the activity of PI3K/Akt signaling pathway through Cav-1.
