摘 要:目的观察蝙蝠葛诺林碱对大鼠皮层神经元氧糖剥夺损伤(OGD)的保护作用及其机制。方法运用噻唑蓝(MTT)、流式细胞技术、二氯二氢荧光素二乙酯DCFH-DA荧光探针和阳离子荧光染色剂JC-1,检测活性氧(ROS)、线粒体膜电位MMP。结果蝙蝠葛诺林碱(0.1,1,10μg·mL^-1)能减轻OGD复灌对大鼠皮层神经元的损伤,有效降低OGD复灌后细胞内ROS的产生及稳定线粒体膜电位。结论蝙蝠葛诺林碱在离体水平显著改善OGD复灌损伤所致的毒性作用,其抗损伤机制与抑制细胞内活性氧生成、稳定线粒体膜电位、保护线粒体功能有关。Objective To observe the protective effect and mechanism of daurinoline on oxygen-glucose deprivation(OGD)in rat cortical neurons.Methods Using MTT,flow cytometry,2,7'-dichlorofluorescein diacetate(DCFH-DA)fluorescent probe,and cationic fluorescent dye JC-1,the reactive oxygen species(ROS)generation and mitochondrial membrane potential(MMP)in rat cortical cultures were detected.Results Daurinoline(0.1,1,10μg·mL^-1)could reduce the injury of OGD reperfusion on the cortical neurons of rats.And it could decrease the intracellular ROS generation and stabilize mitochondrial membrane potential after OGD reperfusion.Conclusion Daurinoline markedly protected rat primary cortical neurons against OGD-reoxygenation-induced toxicity in vitro.The possible mechanism underlying daurinoline protective effects involved regulating ROS generation,stabilizing membrane potential,and protecting mitochondrial function.
