摘 要:目的探讨柚皮素磷脂复合物(NPC)对实验性脉络膜新生血管NF-κB/iNOS通路的影响。方法建立大鼠脉络膜新生血管模型,随机分为空白对照组、溶媒组、柚皮素(Nar)治疗组、NPC高、中、低治疗组,眼底血管荧光造影(FFA)检查分析评估CNV形成率,脉络膜铺片荧光显微镜观察CNV面积,实时荧光定量聚合酶链锁反应(RT-qPCR)和Western blot分别检测眼后节组织中NF-κB、iNOS、COX-2、VEGF、MMP-2、MMP-9 mRNA和蛋白表达。结果FFA检查可见空白对照组,溶媒组显示有强荧光渗漏,荧光素渗透面积分别扩大180点、175点,占总光凝数192点的93.75%、91.14%;Nar组荧光信号减弱,渗透范围扩大161点,占总光凝点数的83.85%,与空白对照组相比CNV形成率差异有统计学意义(X^2=9.454,P=0.000);NPC低、中、高剂量组较空白对照组荧光信号显著减弱,渗透范围分别为143点、120点、105点,分别占总光凝数的74.47%、62.5%、54.68%,光凝4周后CNV形成率差异均有统计学意义(X^2NPC低=26.681、X^2NPC中=54.857、X^2NPC高=76.555,均P=0.000)。治疗4周后,空白对照组、溶媒组新生血管面积最大,两组之间无统计学意义(P>0.05)。与空白对照组相比,Nar组和NPC组CNV面积明显减少,差异有统计学意义(tNar=15.165,tNPC低=21.411,tNPC中=29.815,tNPC高=34.129,均P=0.000);NPC低、中、高三组组间比较总体差异有统计学意义(F=35.117,P=0.000)。NPC高剂量组CNV面积显著小于低、中剂量组(tNPC低=8.275,PNPC低=0.000;tNPC中=3.529,PNPC中=0.002)。与Nar相比,NPC低、中、高剂量组CNV面积明显减少(tNPC低=8.824,tNPC中=13.920,tNPC高=17.724,均P=0.000);与空白对照组相比,溶媒组6项观察指标mRNA表达水平均无统计学意义(P>0.05);Nar组COX-2、MMP-2、MMP-9 mRNA表达水平与空白组相比有统计学意义(tcox-2=10.824,tMMP-2=6.346,tMMP-9=14.710,均P=0.000);NPC低、中、高剂量三组组间在NF-κB、COX-2、VEGF mRNA有统计学意义(FNF-κB=95.681,Fcox-2=65.732,FVEGF=86OBJECTIVE To investigate the effect of naringin phospholipid complex(NPC)on the NF-κB/iNOS pathway of experimental choroidal neovascularization(CNV).METHODS Choroidal neovascularization model of rats was established and randomly divided into blank control group,solvent group,naringenin(Nar)treatment group,NPC high,medium and low doses treatment groups.The FFA was used to evaluate the formation rate of CNV,the area of CNV was observed by choroidal film fluorescence microscopy.The RT-qPCR detected the expressions of NF-κB,iNOS,COX-2,VEGF,MMP-2,and MMP-9 mRNAs in the posterior segment of eye tissues,Western blot was used to detect the expressions of NF-κB,iNOS,COX-2,VEGF,MMP-2 and MMP-9 proteins.RESULTS The FFA showed a strong fluorescence leakage in blank control group and the solvent group.The luciferin penetration area increased by 180 points and 175 points respectively,accounting for 93.75%and 91.14%of the total photocoagulation number of 192 points.The fluorescence signal of naringin group was weakened,and the infiltration range was increased by 161 points,accounting for 83.85%of the total number of light condensation points.Compared with the blank control group,the difference of CNV formation rate was statistically significant(X^2=9.454,P=0.000).Compared with the blank control group,the fluorescence signal of NPC in the low,medium and high dose groups was significantly weakened,and the penetration range was 143 points,120 points and 105 points,respectively,accounting for 74.47%,62.5%and 54.68%of the total photocoagulation number,respectively.The variation in the formation rate of CNV after 4 weeks of photocoagulation was statistically significant(X^2=26.681,54.857,76.555,all P=0.000).After 4 weeks of treatment,there was no statistical significance between the blank control group and solvent group.Compared with the blank control group,the CNV area of naringenin group and NPC group decreased significantly(tnar=15.165,tlow=21.411,tmedium=29.815,thigh=34.129,all P=0.000).The overall difference of NPC among the
