山慈菇提取物对肝癌细胞增殖及TNF-α、IL-1β、IL-6表达的影响

摘　　要：目的观察山慈菇提取物对肝癌SMMC-7721细胞增殖及TNF-α、IL-1β、IL-6表达的影响。方法对SMMC-7721细胞分别给予0、100、200、400、600、800、1000 mg/L山慈菇提取物培养24、48、72 h,采用CCK-8法检测细胞增殖抑制率。将SMMC-7721细胞分为0 mg/L组、400 mg/L组、600 mg/L组、800 mg/L组,分别加入0、400、600、800 mg/L的山慈菇提取物培养24 h,采用平板克隆形成实验测算细胞克隆形成率,光学倒置显微镜下观察细胞形态变化,采用qRT-PCR法检测细胞中的TNF-α、IL-1β、IL-6 mRNA,采用ELISA法检测细胞培养液上清中的TNF-α、IL-1β、IL-6。结果400、600、800 mg/L组细胞克隆形成率低于0 mg/L组,600、800 mg/L组克隆形成率低于400 mg/L组(P均<0.05)。400、600、800 mg/L组随着药物浓度增大,细胞形态皱缩,贴壁差,细胞易脱落,尤以800 mg/L组最明显。400、600、800 mg/L组细胞中TNF-α、IL-1β、IL-6 mRNA表达及细胞培养液上清TNF-α、IL-1β、IL-6水平均低于0 mg/L组,600、800 mg/L组低于400 mg/L组(P均<0.05)。结论山慈菇提取物可抑制肝癌SMMC-7721细胞增殖,并抑制细胞中TNF-α、IL-1β、IL-6的表达与分泌。山慈菇提取物可能是通过抑制炎症因子分泌,调节肿瘤微环境,从而抑制肿瘤细胞增殖。Objective To observe the effects of iphigenia indica extract on the proliferation and the expression levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6)of liver cancer SMMC-7721 cells.Methods SMMC-7721 cells were administered 0,100,200,400,600,800,and 1000 mg/L iphigenia indica extracts for 24,48,and 72 h,respectively,and the cell proliferation inhibition rate was detected by CCK-8.SMMC-7721 cells were divided into 0 mg/L group,400 mg/L group,600 mg/L group,and 800 mg/L group,which were cultured with 0,400,600,and 800 mg/L iphigenia indica extract for 24 h.The plate clone formation assay was used to detect the cell clone formation ability;optical inverted microscope was used to observe the change of cell morphology;quantitative real-time PCR(qRT-PCR)was used to detect TNF-α,IL-1β,and IL-6 mRNA;enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of TNF-α,IL-1β,and IL-6 in the supernatants of cell culture fluid.Results The colony formation rates were lower in the 400,600,and 800 mg/L groups than in the 0 mg/L group,and the colony formation rates were lower in the 600,800 and 800 mg/L groups than in the 400 mg/L group(all P<0.05).With the increase of drug concentrations in the 400,600,and 800 mg/L groups,the cell morphology shrank,adherence was poor,and the cells were prone to fall off,especially in the 800 mg/L group.The expression levels of TNF-α,IL-1β,and IL-6 mRNA and the levels of TNF-α,IL-1β,and IL-6 in cell culture supernatants in the 400,600,and 800 mg/L groups were lower than those in the 0 mg/L group,those in the 600 and 800 mg/L groups were lower than those in the 400 mg/L group(all P<0.05).Conclusions Iphigenia indica extract can inhibit the proliferation of liver cancer SMMC-7721 cells,and inhibit the expression and secretion of TNF-α,IL-1β,and IL-6 in cells.Iphigenia indica extract may inhibit tumor cell proliferation by inhibiting secretion of inflammatory factors and regulating tumor microenvironment.
