摘  要:目的观察不同浓度脱脂牛血清白蛋白(BSA)对人外周血单个核细胞(PBMCs)NOD样受体蛋白3(NLRP3)炎症通路的影响。方法收集健康体检者的外周血,分为BSA组及空白对照组,BSA组分别加入0.3125%、0.625%、1.25%、2.5%、5%、10%脱脂BSA培养6 h,空白对照组加入等量PBS培养6 h。提取各组PBMCs,采用qRT-PCR法检测细胞中的NLRP3炎症通路相关蛋白NLRP3、凋亡相关斑点样蛋白(ASC)、半胱氨酸天冬氨酸蛋白水解酶1(Caspase-1)、白细胞介素1β(IL-1β)mRNA。结果BSA组加入1.25%、2.5%、5%、10%脱脂BSA后,IL-1βmRNA表达水平高于空白对照组(P均<0.05)。BSA组加入1.25%、2.5%、5%、10%脱脂BSA后,ASC mRNA表达水平低于空白对照组(P均<0.05)。BSA组与空白对照组NLRP3、Caspase-1 mRNA表达差异无统计学意义(P均>0.05)。结论1.25%~10%的脱脂BSA刺激PBMCs可上调细胞中IL-1βmRNA表达、下调ASC mRNA表达,对NLRP3、Caspase-1 mRNA表达无显著影响。Objective To observe the effects of different concentrations of defatted bovine serum albumin(BSA)on the inflammatory pathway of NOD-like receptor protein 3(NLRP3)in human peripheral blood mononuclear cells(PBMCs).Methods The peripheral blood of healthy subjects was collected and divided into the BSA group and blank control group.The peripheral blood of the BSA group was added with 0.3125%,0.625%,1.25%,2.5%,5%,and 10%defatted BSA,and the blank control group with equal volume of PBS.The PBMCs were extracted from each group after 6 hours of culture.The expression levels of NLRP3 inflammatory pathway-associated protein NLRP3,apoptosis-related spot-like protein(ASC),Caspase-1,and interleukin 1β(IL-1β)mRNA were detected by real-time fluorescence quantitative PCR(qRT-PCR).Results After adding 1.25%,2.5%,5%and 10%defatted BSA in the BSA group,the IL-1βmRNA expression was significantly higher than that in the blank control group(P<0.05).After adding 1.25%,2.5%,5%and 10%defatted BSA in the BSA group,the expression of ASC mRNA was significantly lower than that in the blank control group(P<0.05).There were no significant differences in NLRP3 or Caspase-1 mRNA expression between the BSA group and the control group(both P>0.05).Conclusion The defatted BSA(1.25%-10%)-stimulated PBMCs can up-regulate IL-1βmRNA expression and down-regulate ASC mRNA expression,but has no significant effect on NLRP3 and Caspase-1 mRNA expression.
