摘 要:目的探讨景虎通脉方对急性脑缺血的治疗作用及机制。方法健康成年雄性Wistar大鼠80只,随机分为假手术对照组、模型对照组、尼莫地平组、景虎通脉方高剂量组、景虎通脉方中剂量组、景虎通脉方低剂量组。每组10只。除去假手术对照组,余各组大鼠采用线栓法制作急性脑缺血动物模型。造模前预防给药7次,造模后给药3次,每天1次。检测大鼠脑组织匀浆中MDA、NOS的含量及SOD的活力。结果与模型组相比,景虎通脉方高、中、低剂量组脑组织匀浆中SOD水平分别为(544.76±40.19)U·mL^(-1)、(537.76±38.80)U·mL^(-1)、(524.43±61.18)U·mL^(-1),景虎通脉方高、中剂量组脑组织匀浆中NOS水平为(37.83±4.36)U·mL^(-1)、(36.94±2.18)U·mL^(-1),景虎通脉方中剂量组脑组织匀浆中MDA水平为(3.36±1.55)U·mL^(-1),结果具有统计学意义。结论景虎通脉方对大鼠急性脑缺血有一定的治疗作用,其机制可能与抗氧自由基及抗氧化途径有关。Objective:To explore the therapeutic effect and mechanism of Jinghu Tongmai Recipe on acute cerebral ischemia.Methods:80 healthy adult male Wistar rats were randomly divided into a sham operation control group,a model control group,a nimodipine group,and Jinghu Tongmai Recipe groups with high,medium and low dose,10 rats per group.Except the rats in the sham operation control group,the rats in the other groups were made into animal models of acute cerebral ischemia by suture method.Preventive administration was given 7 times before modeling and 3 times after modeling,once a day.The contents of MDA and NOS and the activity of SOD in rat brain tissue homogenate were detected.Results:Compared with that of the model group,the levels of SOD in brain tissue homogenates of Jinghu Tongmai Recipe groups with high,medium and low dose were(544.76±40.19)U·mL-1,(537.76±38.80)U·mL-1 and(524.43±61.18)U·mL-1,respectively.The NOS levels in brain tissue homogenates of the high and medium dose groups were(37.83±4.36)U·mL-1 and(36.94±2.18)U·mL-1,The MDA level in the brain tissue homogenate of the medium dose group was(3.36±1.55)U·mL-1.The results were statistically significant.Conclusion:Jinghu Tongmai Recipe has a certain therapeutic effect on acute cerebral ischemia in rats,and its mechanism may be related to antioxidant free radicals and antioxidant pathways.
