摘 要:选用未破壁的松花粉作为样品,通过显微镜观察法优化松花粉的悬浮分散效果,筛选50g︰20m L︰15m L比例混合的水合氯醛-水-甘油溶液作为悬浮液、涡旋时间为60 s和放置时间为90 min时载玻片上的松花粉分散效果最佳。并通过破壁料理机对松花粉进行破壁处理,采用响应面方法对影响破壁率的3个主要因素(样品量、单次运行时间和循环次数)进行优化。结果表明:当样品量40 mg,单次运行时间90 s,循环次数5次时,松花粉的破壁率可达到96.63%。方差分析结果表明,影响破壁料理机破壁松花粉的因素由强到弱依次为:单次运行时间>循环次数>样品量。The unbroken pine pollen was used as a sample to optimize the suspension and dispersion effect of pine pollen by microscopic observation. When the mixed chloral hydrate-water-glycerol solution with 50 g︰20 mL︰15 mL ratio was selected as suspension, the vortex time was 60 s and the standing time was 90 min,the dispersion effect of pine pollen on slide was the best. The wall-breaking treatment of pine pollen was carried out by using a wall-breaking material machine, the main factors influencing the broken wall process was as follows: the sample volume, the single run time and the cycle index was optimized. The results shows that the breaking rate of pine pollen reached 96.63 %, when the sample volume was 40 mg, the single run time was 90 s and the cycle index was 5 times. The results of variance analysis showed that the factors affecting broken wall process was extruded from strong to weak: single run time>cycle index>sample volume.
