黄连解毒汤含药血清对泡沫细胞ABCA1表达与胆固醇含量的影响

摘　　要：目的:观察黄连解毒汤含药血清对泡沫细胞中三磷酸腺苷结合盒转运体A1(ABCA1)表达及胆固醇含量的影响,探讨黄连解毒汤抗动脉粥样硬化的可能作用机制。方法:SD大鼠给予大、中、小剂量黄连解毒汤灌胃后制备含药血清。用氧化低密度脂蛋白(ox-LDL)诱导RAW264.7巨噬细胞分化为泡沫细胞,将细胞分为空白组、模型组、黄连解毒汤含药血清大剂量组、中剂量组、小剂量组,每组1.5×10^6个细胞,分别加入无血清培养液和不同浓度含药血清干预24 h,总胆固醇及游离胆固醇试剂盒检测细胞中胆固醇含量,实时荧光定量PCR检测细胞ABCA1 mRNA的表达。结果:与模型组比较,黄连解毒汤含药血清各剂量组泡沫细胞内总胆固醇(TC)含量明显降低(P<0.05或P<0.01),大剂量组游离胆固醇(FC)含量显著下降(P<0.01);含药血清各剂量组ABCA1 mRNA表达均明显升高(P<0.05)。结论:黄连解毒汤上调泡沫细胞ABCA1的转录与表达,促进胆固醇流出,可能是其抗动脉粥样硬化的机制之一。Objective To observe the effect of serum containing Huanglian Jiedu Decoction on the expression of ATP-binding cassette transporter(ABCA1)and cholesterol content in foam cells.To explore the possible mechanism of action of Huanglian Jiedu Decoction against atherosclerosis.Methods SD rats were given Large,medium and small doses of Huanglian Jiedu Decoction to prepare drug-containing serum.RAW264.7 macrophages were induced to differentiate into foam cells by oxidized low density lipoprotein(ox-LDL).The cells were divided into control group,model group,Huanglian Jiedu Decoction-containing serum high-dose group,medium-dose group,and low-dose group.1.5×10^6 cells in each group were added to serum-free medium and different concentrations of drug-containing serum for 24h.The total cholesterol(TC)and free cholesterol(FC)kits were used to detect the cholesterol content in the cells,and the expression of ABCA1 mRNA was detected by quantitative real-time PCR.Result Compared with the model group,TC content in the foam cells of Huanglian Jiedu Decoction group was significantly decreased(P<0.01),and the FC content in the high dose group was significantly decreased(P<0.01).The expression of ABCA1 mRNA in serum dose group was significantly increased(P<0.05).Conclusion Huanglian Jiedu Decoction up-regulates the transcription and expression of foam cell ABCA1 and promotes cholesterol efflux,which may be one of the mechanisms of its anti-atherosclerosis.
