摘 要:针对水库生态调度及其生态影响的问题,在2018年6月汉江中下游丹江口、崔家营、兴隆等梯级水库联合生态调度期间,依托汉江下游仙桃水文站布设断面,开展水环境监测及采样、测试分析,研究水库生态调度过程中河流细菌群落的变化特征。利用YSI水质参数仪现场测定水温、pH值、溶解氧、电导率,依据分光光度法测定总氮、总磷,采用高通量测序方法分析细菌群落多样性、丰富度及非度量多维尺度分析法分析细菌群落结构的变化情况。结果表明,水库生态调度过程中下游水位显著上升,流量显著增大,pH值与电导率显著下降,水温、溶解氧、总氮、总磷未发生显著变化;细菌群落多样性与丰富度提高,结构相似性降低;β变形菌门相对丰度明显增加,疣微菌门相对丰度明显降低。该结论为水库生态调度提供了参考依据。Aiming at the problem of reservoir ecological dispatch and its ecological impact,based on the layout section of Xiantao hydrometric station in the lower reaches of the Hanjiang River,water environment monitoring,sampling,testing and analyzing were carried out to study the characteristics of variation in fluvial bacterial communities during the reservoirs ecological regulation of Danjiangkou,Cuijiaying and Xinglong in the middle and lower reaches of the Hanjiang River in June 2018.The YSI water quality parameter meter was used to measure water temperature,pH,dissolved oxygen and conductivity.Total nitrogen and total phosphorus were determined by spectrophotometry.The diversity and richness of bacterial community were analyzed by high-throughput sequencing method,and the changes of bacterial community structure were analyzed by non-metric multidimensional scaling analysis.During the ecological dispatching process,the water level in the lower reaches of the reservoir increased significantly,the discharge rate increased significantly,the pH and conductivity decreased significantly,but there were no obvious changes in water temperature,dissolved oxygen,total nitrogen and total phosphorushad.There is an increase in the diversity and richness of the bacterial communities and a decrease in the structural similarity.The relative abundance of Betaproteobacteria was significantly increased,and the relative abundance of Verrucomicrobia was significantly reduced.This paper provides a reference for the ecological regulation of reservoirs.
