摘  要:为了研究MADS-box E类基因在荷花花器官发育中的作用,以荷花‘重瓣一丈青’(Nelumbo nucifera'Chongban yizhangqing')品种为试验材料,利用RT-PCR克隆获得AGL6-like基因,命名为NnMADS6。序列分析表明,NnMADS6基因全长770 bp,含有1个726 bp的开放读码框(ORF),编码241个氨基酸,C-末端包含典型SEP/AGL6基序。氨基酸序列比对发现Nn MADS6与红花木兰MawAGL6、腊梅ChpAGL6、黑种草NidAGL6、无油樟AmtMADS6、烟草NitMADS6和拟南芥AtAGL6同源性分别为83.5%、81.5%、81.2%、80.9%、73.4%和59.1%,进一步系统进化树分析显示NnMADS6基因聚在AGL6分支中基本被子植物类群,说明该基因属于MADS-box家族AP1/AGL9亚家族AGL6分支同源基因。表达模式分析显示,无论在单瓣型材料或重瓣型材料,NnMADS6基因主要集中在花芽、萼片、花瓣和心皮中表达,花瓣中表达量最高。本研究结果表明,NnMADS6基因在花器官发育尤其在花瓣形成中有调控作用。To study the role of MADS-box E gene in the flower organs development of Nelumbo nucifera,flower buds of Nelumbo nucifera’Chongban yizhangqing’cultivars were used as tested materials and a AGL6-like gene named as NnMADS6 was cloned by RT-PCR.Sequence analysis showed that full length of the NnMADS6 gene was 770 bp,containing an open reading frame(ORF)of 726 bp,encoding 241 amino acids,and C-terminal contained a typical SEP/AGL6 motif.Amino acid sequence alignment showed that NnMADS6 had 83.5%,81.5%,81.2%,80.9%,73.4%and 59.1%homologies with MawAGL6(Magnolia wufengensis),ChpAGL6(Chimonanthus praecox),NidAGL6(Nigella damascena),AmtMADS6(Amborella trichopoda),NitMADS6(Nicotiana tabacum)and AtAGL6(Arabidopsis thaliana),respectively.Further phylogenetic tree analysis showed that the NnMADS6 gene was clustered in the basic angiosperm taxa of AGL6 branch,indicating that the gene obtained in this study belonged to the AGL6 branch homology gene of AP1/AGL9 subfamily of MADS-box family.Expression pattern analysis showed that NnMADS6 gene was mainly expressed in bud,sepal,petal and carpel and the highest expression was found in petals,regardless of single-petalled and double-petalled cultivars in lotus.The results of this study showed that,NnMADS6 gene plays a regulatory role in floral organ development,especially in petal formation.
