摘  要:为探明低温胁迫处理对不同甘蔗转录因子表达的影响,以‘桂糖08-1180’和‘ROC22’为试验材料,进行低温和常温处理,经转录组测序后,对转录因子基因的差异表达进行了分析。结果表明:测序共获得360404条Transcripts,183515个Unigenes。低温胁迫下‘桂糖08-1180’获得了566个差异表达转录因子基因,隶属于61类转录因子家族。‘ROC22’获得了769个差异表达转录因子基因,隶属于71类转录因子家族。2个品种的差异表达转录因子基因表现为上调表达基因数多于下调表达基因数。ROC22在MYB、AP2-EREBP、GRAS、WRKY、NAC、Orphans、bZIP、FAR1、C3H、C2H2等家族中具有较多的转录因子基因:‘桂糖08-1180’在AP2-EREBP、WRKY、MYB、Orphans、b ZIP、NAC、GRAS、C2H2、FAR1、bHLH等家族中具有较多转录因子基因。有60个转录因子家族包含了2个品种的差异表达转录因子基因,而BES1、CAMTA、CPP等仅含有‘ROC22’的差异表达转录因子基因,CSD仅含有‘桂糖08-1180’的差异表达转录因子基因。从差异表达倍数来看,2个品种中均以表达倍数在10倍以下的转录因子基因最多。在‘ROC22’中,差异倍数较大的转录因子主要分布在AP2-EREBP、Tify和WRKY家族,在‘桂糖08-1180’中,差异倍数较大的转录因子主要分布在MYB、WRKY和AP2-EREBP家族。本研究为甘蔗抗寒相关转录因子的挖掘及甘蔗抗寒性机理研究提供帮助。In order to find out the effect of cold stress treatment on expression of transcription factor in different sugarcane,both’GT08-1180’and’ROC22’under cold stress treatment and normal temperature treatment were sequenced with transcriptome sequencing technologies,and different expression of transcription factor genes were analyzed.The results showed that 360404 transcripts and 183515 unigenes were obtained,and 566 differentially expressed transcription factors(TFs)belonging to 61 families were found in’GT08-1180’,769 differentially expressed TFs belonging to 71 families were found in’ROC22’.In addition,there were more up-regulated TFs than down-regulated TFs in both’GT08-1180’and ROC22.MYB,AP2-EREBP,GRAS,WRKY,NAC,Orphans,bZIP,FAR1,C3 H and C2 H2 were the largest number families of differentially expressed trancription factors in’ROC22’,then AP2-EREBP,WRKY,MYB,Orphans,b ZIP,NAC,GRAS,C2 H2,FAR1 and bHLH were the largest number families of differentially expressed trancription factors in’GT08-1180’.Total 60 trancription factor families had the same differentially expressed trancription factors genes in both’GT08-1180’and’ROC22’,while BES1,CAMTA,CPP,DBP,HRT,LIM,PLATZ,Rcd1-like,S1 Fa-like,TIG and VOZ had the special differentially expressed trancription factors genes only in’ROC22’,CSD had the special differentially expressed trancription factors genes only in’GT08-1180’.Fold change(FC)values of the most TFs were less than 10 in 2 cultivars.It was found that AP2-EREBP,Tify and WRKY had higher FC values in’ROC22’,but MYB,WRKY and AP2-EREBP had higher FC values in’GT08-1180’.In this paper,it is provided the foundation in transcription factors of cold resistance and the mechanism of cold resistance in sugarcane.
