摘 要:CMK是MEP途径中的第四个关键酶,对于萜类合成代谢具有重要的影响。本研究基于实验室前期获得的山胡椒果实的转录组数据,经过注释筛选出CMK基因,首次从山胡椒中克隆得到萜类生物合成关键基因LgCMK并对其生物信息学预测分析。结果显示:LgCMK基因的开放阅读框为1 221 bp,编码406个氨基酸,分子量为44.78 k D,理论上等电点为6.26,属GHMP蛋白超家族。对Lg CMK蛋白的氨基酸序列理化性质进行分析,发现蛋白亲水性指数为-0.332,不稳定性指数为41.77,属于亲水性不稳定蛋白;亚细胞定位预测结果显示该蛋白定位于叶绿体中。同源性和系统进化分析结果表明,与香樟、博落回和罂粟同源蛋白相似度较高分别为96.8%、75.96%和73.0%,不同物种的CMK具有较近的亲缘关系,与香樟同属一支,亲缘关系最近。通过本研究生物信息学分析将有助于对LgCMK深层次功能探究提供借鉴作用,同时为山胡椒的种质改良提供基因资源。CMK is the fourth key enzyme in the MEP pathway and has important implications for steroidal anabolism.Based on the transcriptome data of the Lindera glauca fruit obtained in the early stage of the laboratory,the CMK gene was screened by annotation,and the key gene of steroid biosynthesis LgCMK was cloned from Lindera glauca for the first time and its bioinformatics prediction analysis was carried out.The results showed that the open reading frame of LgCMK gene was 1221 bp,encoding 406 amino acids with a molecular weight of 44.78 kD and a theoretical isoelectric point of 6.26,belonging to the GHMP protein superfamily.The physicochemical properties of the amino acid sequence of the LgCMK protein were found to be-0.332 and the instability index was 41.77,which was a hydrophilic unstable protein;the subcellular localization prediction showed that the protein was localized in the chloroplast.The results of homology and phylogenetic analysis showed that the similarities with the Cinnamomum camphora,Macleaya cordata and Papaver somniferum homologous proteins were 96.8%,75.96%and 73.0%,respectively,and the CMK of different species had close kinship.It belongs to the same family and has a close relationship.Through the bioinformatics analysis of this study,it will help to provide a reference for the deep functional exploration of LgCMK,and provide genetic resources for the germplasm improvement of Lindera glauca.
