摘 要:花叶凤尾竹(Bambusa glaucescens f.albo-variegata)具有很高的观赏价值,但现存量少,需通过组织培养技术实现快速繁殖的目标。本研究以花叶凤尾竹当年生幼嫩带节茎段为外植体进行组织培养,探索花叶凤尾竹组织培养外植体采集时间、消毒条件、增殖和生根的最佳培养基配方。结果表明,以6月下旬采集保留叶鞘的花叶凤尾竹幼嫩带节茎段为外植体,可得到最高的存活率;最佳消毒方式为有效氯浓度1%的NaClO溶液,消毒12 min;最佳丛芽诱导和增殖的培养基为MS+0.08 mg/L TDZ+1 mg/L BA+0.2 mg/L KT;生根培养基为1/2MS+2.5 mg/L NAA。综上建立的花叶凤尾竹快繁体系,无菌苗获得率可达58.88%,增殖系数可达3.65,生根率86.67%,平均生根数3以上。本研究为快速繁育花叶凤尾竹种苗提供技术支持。Bambusa glaucescens f.albo-variegata has very high ornamental values,but the stock of this bamboo species is quite limited,resulting in an urgent need of study on tissue culture to achieve its rapid reproduction.In this study,the young stems of Bambusa glaucescens f.albo-variegata with axillary buds were used for the in-vitromicropropagation.Our main objectives were to explore the optimal scheme of the explant obtaining time,sterilization condition,growth regulator concentration applicated for proliferation and rooting.The results showed that the highest survival was observed in the treatment which explants were obtained in late June,and the best disinfection method was 1%of NaClO solution for twelve minutes.Concomitantly,the MS medium supplemented with 0.08 mg/L TDZ,1 mg/L 6-BA,and 0.2 mg/L KT was optimal for shoot multiplication,while the 1/2 MS medium supplemented with 2.5 mg/L of NAA was optimal for rooting.With the rapid propagation regimes stated,we yielded 58.88%of aseptic plantlets and the proliferation coefficient reached 3.65;a 86.67%of rooting rate was observed and the average rooting number was more than three.Our results will provide technical support for the rapid propagation of the seedlings of Bambusa glaucescens f.albo-variegata.
