摘 要:龙须菜是一种重要的产琼胶海藻,对其繁殖发育的研究将有助于了解龙须菜的生长,有助于育种。本研究对龙须菜不同育性品系(981和鲁龙1号)在不同发育阶段的表达谱数据,采用加权基因共表达网络分析(WGCNA)方法构建基因共表达网络,共获得32个模块。其中,模块最大基因数量为2577个,最小基因数为66个。经过筛选发现与品系相关的模块(以0.7为阈值)共5个,涉及基因共2301个,对模块基因进行GO功能富集分析,结果表明主要富集以下功能上:信号转导、转运、细胞分化、翻译、细胞稳态、生物合成过程等。981品系与鲁龙1号品系的富集结果对比表明981品系在孢子放散过程中生物活性不高,生物合成水平较低,内环境稳态较差。经过KEGG富集,筛选到21个与981品系II期四分孢子形成相关的备选基因,主要富集到DNA复制和染色体形态变化等与细胞周期相关的功能上,表明981品系在孢子形成时期的细胞分裂可能出现异常。本文结果为进一步深入揭示龙须菜四分孢子体发育分子调控机制提供思路和导向。Gracilariopsis lemaneiformis is an important agarophyta.Stydies on its reproduction and development will help to understand the growth of G.lemaneiformis and be applicable in breeding.In this study,the digital gene expression(DGE)profiles of different fertility cultivar(981 and Lulong No.1)in different developmental stages were used to construct a gene co-expression network using WGCNA and a total of 32 modules were obtained.Gene numbers of modules are ranged from 66 to 2577.After screening,five modules related to the cultivar(with 0.7 as the threshold)were found,involving a total of 2301 genes.The GO function enrichment analysis of the module genes showed that the main functions were:signal transduction,transport,cell differentiation,translation,cell homeostasis,biosynthesis processes,et al.The comparison of the enrichment results between cultivar 981 and Lulong No.1 further indicated thatcultivar 981 had low biological activity,low biosynthesis level and poor homeostasis during the tetraspore release process.In addition,21 candidate genes related to the tetraspore formation of cultivar 981 were screened after KEGG enrichment.They mainly enriched on cell cycle-related functions such as DNA replication and chromosome morphological changes,which suggesting that cultivar 981 is abnormal in cell division during the sporulation period.This study provided ideas and guidance for further revealing the molecular regulation mechanism of the development of the tetrasporohyte of G.lemaneiformis.
