奶山羊发情期与间情期卵巢中CircRNA差异性表达谱分析

摘  要:circRNAs参与许多重要的生物过程,并作为miRNA的“分子海绵”发挥重要作用。目前关于卵巢中circRNA的种类、数量、功能及其对卵巢发育的调控机理研究报道很少。本研究以发情期与间情期的奶山羊卵巢为研究对象,通过样品提取总RNA后并对其进行处理之后制备整个文库的策略,利用IlluminaHiSeq进行高通量测序并进行分析、生物信息学方法,采用RT-qPCR等检测技术,研究了奶山羊发情期与间情期卵巢组织中差异的circRNA表达谱。结果表明,奶山羊发情期和发情间期卵巢组织中候选的circRNA有22,333个,可分为六类;奶山羊发情期卵巢相对发情间期有772个circRNA差异性表达,268个上调,504个下调;奶山羊卵巢组织中除了Y染色体,circRNAs在其他所有染色体上均广泛分布,大多数circRNA长度约为500nt。因此本研究对奶山羊发情期和发情间期卵巢组织中circRNA表达谱进行检测和分析,为进一步探索circRNA在奶山羊发情周期卵巢发育中的调控机理提供试验依据,也为提高奶山羊的繁殖率提供新的思路与方法。CircRNAs are involved in many important biological processes and play an important role as"molecular sponges"for miRNAs.At present,there are few reports on the type,function and regulatory mechanism of circRNA in ovarian development.In this study,the ovaries of the estrus and interstitial cows were used as the research object.The whole library was prepared by extracting total RNA from the sample and processing it.The Illumina HiSeq for sequencing and analysis and RT-qPCR techniques were used,the circRNA in ovaries of milk goats during estrus and interestrus stage and its role in the estrus process of milk goats were studied.The results showed that there were 22,333 candidate circRNAs in ovarian tissue of estrus and estrus,which could be divided into six categories.There were 772 differential circRNAs in ovary relative estrus of dairy goats,among them,268 up-regulated expression,504 down-regulated;in addition to the Y chromosome in ovary tissue of dairy goats,circRNAs were widely distributed on all other chromosomes,and most circRNAs were approximately 500nt in length.In summary,this study investigated the circRNA expression in ovarian tissue of dairy goats at different developmental stages.It opens up a new field for the function of circRNA in ovarian development and provides data support for the specific biological functions and molecular mechanisms of circRNA involved in ovarian development and estrus in dairy goats.
