摘 要:某养殖场幼龄蓝狐(Alopex lagopus)不同程度脱水、生长停滞,剖检可见肾脏苍白肿大。取肾脏、脑组织研磨,差速离心后染色检测到典型脑炎原虫形态,大小1~2μm。肾脏石蜡切片经革兰染色、改良马松三色染色观察到大量染色虫体。肾脏研磨液提取基因组DNA,根据NCBI的脑炎原虫(Encephalitozoon cuniculi,E.cuniculi)基因组设计2对特异性引物,分别扩增脑炎原虫核糖体DNA内转录间隔区1(ITS1)和极管蛋白1(PTP1)的基因片段。PCR结果显示,引物ITS1和PTP1分别扩增出762 bp和1 071 bp的片段,两片段序列与脑炎原虫相应序列的相似性均为99.7%。对ITS1基因序列进行分析,鉴定为脑炎原虫基因Ⅲ型。此外,PTP1基因的系统进化树分析了该菌株(称为JL-1),与2014年辽宁LN-1株、2001年美国CDC. V282株关系最近。Blue fox encephalitozoonosis is a chronic or recessive parasitic disease caused by Encephalitozoon cuniculi.Blue fox is one of the most susceptible species to Encephalitozoon cuniculi.Stunting,blindness,pale and swollen kidneys and vertical transmission are the main features of the disease.In 2017,an outbreak of blue fox disease occurred in Jilin Province(China),and caused a widespread infection in bred young foxes.The typical morphology of encephalitozoonosis was observed in the kidney by smear Gram staining and modified masson trichrome staining.A typical granulomatous lesion was observed in the pathological section.It was identified as type Ⅲ of Encephalitozoon cuniculi gene by ITS1 gene sequence analysis.In addition,the phylogenetic tree of the PTP1 gene of this strain was analyzed(referred to as JL-1),and the data showod that this strain was closely related to the 2014 LN-1 strain in Iiaoning.
