摘  要:目的:评价消癌平注射液对斑马鱼卵巢癌移植瘤的抑制作用。方法:采用CM-DiI荧光标记的人卵巢癌A2780细胞,以显微注射的方式移植到斑马鱼卵黄囊内,建立斑马鱼卵巢癌移植瘤模型。根据斑马鱼毒性反应和死亡情况确定正常斑马鱼和荷瘤斑马鱼对消癌平注射液的最大耐受量。将荷瘤斑马鱼分为模型对照组、溶剂对照组、紫杉醇组和消癌平低、中、高剂量组,各组给予相应干预。荧光显微镜测定斑马鱼移植瘤的荧光强度及肿瘤细胞在斑马鱼尾部的转移距离,观察消癌平注射液对斑马鱼移植瘤生长和转移的影响,计算肿瘤生长抑制率和肿瘤转移抑制率。结果:①最大耐受量实验显示,消癌平注射液对正常斑马鱼的毒性较低;当其剂量≤1.25 ng/尾时,荷瘤斑马鱼未出现死亡和毒性反应,故选取0.312 5、0.625和1.25 ng/尾分别作为消癌平注射液低、中、高剂量进行后续试验。②与模型对照组相比,溶剂对照组、紫杉醇组和消癌平低、中、高剂量组的肿瘤生长抑制率分别为3%、33%、15%、14%、27%;紫杉醇组和消癌平高剂量组斑马鱼的整体荧光强度显著降低(P<0.05)。③与模型对照组相比,溶剂对照组、紫杉醇组和消癌平低、中、高剂量组的肿瘤转移抑制率分别为15%、61%、11%、36%、42%;紫杉醇组和消癌平中、高剂量组的肿瘤转移累计距离明显减少(P<0.01)。结论:消癌平注射液对斑马鱼卵巢癌移植瘤的生长和转移均具有一定的抑制作用。Objective:To evaluate the inhibitory effects of Xiao-Ai-Ping Injection on xenografted ovarian tumor in zebrafish. Methods:Human ovarian cancer A2780 cells labeled with CM-DiI were transplanted into zebrafish yolk sac by microinjection to establish the ovarian cancer transplantation model in zebrafish. According to the toxicity and death of zebrafish, the maximum tolerance dose of Xiao-Ai-Ping Injection in normal zebrafish and tumor-bearing zebrafish was determined. The tumor-bearing zebrafish were divided into the model control group, solvent control group, paclitaxel group and Xiao-Ai-Ping groups with low-, medium-and high-dose. Each group was treated with the corresponding intervention. The fluorescence intensity and the metastasis distance of tumor cells in the tail of zebrafish were detected by fluorescence microscope,the effects of Xiao-Ai-Ping Injection on the growth and metastasis of transplanted tumor in zebrafish were observed,and the tumor growth inhibition rate and tumor metastasis inhibition rate were calculated. Results:(1) The maximum tolerance dose test showed that,Xiao-Ai-Ping Injection showed little toxicity on normal zebrafish;at the dose of ≤1.25 ng/tail,the tumor-bearing zebrafish did not show death and toxicity,so 0.312 5,0.625 and 1.25 ng/tail were selected as low-,medium-and highdose for the following test.(2) Compared with the model control group,the tumor growth inhibition rates of the solvent control group,paclitaxel group and Xiao-Ai-Ping groups with low-,medium-and high-dose were 3%,33%,15%,14% and 27%,respectively. The overall fluorescence intensity of zebrafish in the paclitaxel group and Xiao-Ai-Ping group with high-dose was significantly lower than that in the model control group( P<0.05).(3) Compared with the model control group,the inhibition rates of tumor metastasis in the solvent control group,paclitaxel group and Xiao-Ai-Ping groups with low-,medium-and high-dose were 15%,61%,11%,36% and 42%,respectively.
