摘  要:目的对中药显微定量法进行深入的研究。旨在利用星点设计-效应面优化法替换目前国内常用的均匀设计法优化中药显微特征常数测定条件,进而对中药全蝎进行质量评价。方法对全蝎采用刚毛作为显微特征,利用星点设计-效应面优化法优化全蝎刚毛的显微特征常数测定条件,用SPSS 19.0进行二元线性拟合,用OriginPro 9进行二元非线性拟合和响应面的3D函数图绘制。用得到的优化条件测定不同药店全蝎粉末中刚毛的混匀度和显微特征常数数值,用结果与预测值偏差作为本试验方法可行性分析。结果利用星点设计建立的数学模型得到的预测值与结果非常接近,全蝎的显微特征常数混匀度与全蝎用量和甘油量有显著的相关性。结论本试验利用星点设计-效应面优化法建立数学模型,预测性好,显著性高,具有很高的可信度,为中药的显微特征常数测定条件优化及质量评价提供了新的技术手段。Objective To study the micro-quantitative method of traditional Chinese medicine in-depth.The aim was to use the central composite design-response surface methodology to replace the currently used domestic uniform design method to optimize the determination conditions of the microscopic characteristic constants of traditional Chinese medicines.Methods Bristles were used as the microscopic features of the whole gills.Central composite design-response surface methodology was used to optimize the microscopic characteristic constants of the full gill bristles.The SPSS 19.0 was used for the binary linear fitting and the OriginPro 9 was used for binary non-linearity.Linear fitting and 3D function plotting of response surfaces.The optimized conditions were used to determine the degree of bristling mixing and microscopic characteristic constants in bristles powders from different pharmacies.The deviation of the results from the predicted values was used as the feasibility analysis of the experimental method.Results The predicted value obtained by the mathematical model established by the central composite design was very close to the result.The microscopic characteristic constant mixing degree of the whole pupa had a significant correlation with the amount of total hydrazine and glycerol.Conclusion This experiment used the central composite design-response surface methodologytoestablish a mathematical model with good predictability,high significance and high credibility.It provides new technical means for optimizing the determination conditions of microscopic characteristic constants of traditional Chinese medicine.
