摘 要:蛋白质作为生命活动的执行者,其种类和含量的变化在调控生命过程中发挥着至关重要的作用。如何实现深度覆盖的蛋白质组定性定量分析已成为当前蛋白质科学及其相关领域亟需解决的关键问题之一。中国科学院大连化学物理研究所蛋白质组分析新方法研究团队,在科技部重大研究计划、重点研发计划等项目的支持下,针对目前规模化蛋白质分析中存在的精准度和覆盖度有待提高的关键科学问题,以色谱-质谱为核心,发展了高效样品制备新方法、高通量分析新技术和精准定量新方法,取得了一系列创新性成果,显著提高了蛋白质组分析的精准度、覆盖率和分析通量,为我国蛋白质科学研究的深入发展,为推动生物医药、国家安全和人口健康等领域的不断进步提供了重要的技术支撑。As the executors of life activities, proteins play important roles in regulating various biological procedures. Therefore, how to achieve the deep coverage qualitative and quantitative analysis of proteomes has been regarded as one of the key scientific problems in protein science and related fields. With the financial support from National Key Research and Development Program of China and National Basic Research Program of China, the research groups in Dalian Institute of Chemical Physics, Chinese Academy of Sciences, have been devoting into the development of new techniques and methods for high efficiency sample preparation, high resolution chromatography separation, high accuracy quantitation and integrated analysis platforms, by which the coverage, accuracy, precision and throughput of proteome analysis has been obviously improved, and further provide powerful technique supports to solve the bottleneck problems in various fields, such as molecular biology, biopharmaceuticals, public safety and population health.
