摘 要:目的:探索不同剂量五福饮对大鼠尾椎间盘退变(IVDD)髓核β-连环素(β-catenin)、聚蛋白多糖(Aggrecan)、Ⅱ型胶原(Col2)的调控作用。方法:取SD大鼠64只,随机分为空白组、模型组、五福饮低剂量组及五福饮高剂量组。除空白组外,其余各组大鼠构建IVDD模型。模型构建成功后,五福饮低、高剂量组灌服不同剂量的五福饮药液,空白组和模型组大鼠灌服生理盐水。五福饮干预8周后,检测椎间盘髓核β-catenin阳性表达细胞数及mRNA表达水平,Aggrecan及Col2 mRNA水平。结果:模型制备4周后,与空白组比较,模型组、五福饮低、高剂量组大鼠50%缩足阈值(PWT)明显降低(P<0.05);尾椎间盘髓核β-catenin阳性表达细胞数明显升高(P<0.05)。五福饮干预8周后,与空白组比较,模型组大鼠尾椎5~6椎间盘Aggrecan与Col2 mRNA表达明显降低(P<0.05),β-catenin mRNA及阳性表达细胞数明显升高(P<0.05)。与模型组比较,五福饮低、高剂量组大鼠50%PWT、尾椎5~6椎间盘Aggrecan与Col2 mRNA表达均明显升高(P<0.05),β-catenin mRNA及阳性表达细胞数明显降低(P<0.05)。与五福饮低剂量组比较,五福饮高剂量组大鼠50%PWT、尾椎5~6椎间盘Aggrecan与Col2mRNA表达均明显升高(P<0.05),β-catenin mRNA及阳性表达细胞数明显下降(P<0.05)。结论:五福饮可延缓或部分逆转大鼠尾IVDD,且五福饮对大鼠尾IVDD治疗作用存在剂量依赖性。椎间盘髓核β-catenin可能是五福饮的作用靶点。Objective:To explore the regulative effect of different dose of Wufu Yin onβ-catenin in nucleus pulposus,Aggrecan and typeⅡ collagen(Col2) in rats with intervertebral disc degeneration(IVDD) at caudal spine.Methods:A total of 64 SD rats were randomly divided into the blank group,the model group,the low-dose Wufu Yin group and the high-dose Wufu Yin group. IVDD model was constructed in all groups except the blank group. After the model was successfully constructed,the low-dose and high-dose Wufu Yin groups were given different doses of Wufu Yin,while the rats in the blank group and the model group were given normal saline. After eight weeks of intervention with Wufu Yin,the number of positive cells and mRNA expression of β-catenin,and mRNA levels of Aggrecan and Col2 were detected.Results:After four weeks of model preparation,compared with that in the blank group,the levels of 50% paw withdrawal threshold(PWT) in the model group and the low-dose and high-dose Wufu Yin groups were significantly reduced(P<0.05);the number of positive cells of β-catenin in nucleus pulposus of caudal disc was increased significantly(P<0.05). After the eight-week intervention with Wufu Yin,compared with those in the blank group,the mRNA expressions of Aggrecan and Col2 in the fifth to sixth intervertebral discs of rat caudal vertebra in the model group were significantly decreased in the model group(P<0.05);the mRNA expressions of β-catenin and the number of positive cells were significantly increased(P<0.05). Compared with that in the model group,the 50%PWT and the mRNA expressions of Aggrecan and Col2 in the fifth to sixth intervertebral discs of caudal vertebra in the the low-dose and high-dose Wufu Yin groups were significantly increased(P<0.05),and the mRNA expressions of β-catenin and the number of positive cells were significantly decreased(P<0.05).
