摘 要:目的 研究链霉菌Streptomyces sp. FXJ1.088中具有拮抗耐药表皮葡萄球菌活性的次级代谢产物。方法 发酵液和菌丝体通过硅胶柱层析、葡聚糖凝胶柱层析和高效液相色谱等方法分离,化合物结构经质谱、核磁共振、红外光谱等技术鉴定,抗菌活性最低抑菌浓度(minimal inhibitory concentration, MIC)测定采用对倍稀释法。结果 从链霉菌Streptomyces sp.FXJ1.088中分离鉴定了5个化合物,结合文献数据对比,鉴定其结构分别为:antibiotic WS 1627C(A)、antibiotic WS 1627A(B)、epihygromycin(C)、hygromycin A(D)和copiamycin(E)。结论 从链霉菌Streptomyces sp. FXJ 1.088中得到5个化合物,均具有一定的抗多剂耐药表皮葡萄球菌活性。化合物A^E的MIC值分别为100、100、25和25μg/mL以及100~200μg/mL。Objective To investigate secondary metabolites with antagonistic activity against Staphylococcus epidermidis from Streptomyces sp.FXJ1.088.Methods The fermentation broth and mycelium were separated by silica gel column chromatography,dextran gel column chromatography,and high performance liquid chromatography.Compounds were identified by mass spectrometry,nuclear magnetic resonance,and infrared spectroscopy.Antimicrobial bioactivities(MICs)were measured by the double dilution method.Results Five compounds were isolated from the fermentation broth and mycelium of Streptomyces sp.FXJ1.088.By comparing with literature data,their structures were identified as:Antibiotic WS 1627C(A),antibiotic WS 1627A(B),epihygromycin(C),hygromycin A(D),and copiamycin(E).Conclusion Five compounds were obtained from the fermentation broth and mycelium of Streptomyces sp.FXJ1.088.All five compounds showed inhibitory activity against multi-drug resistant S.epidermidis,wherein the MICs of compounds A^E were 100,100,25,25,and 100~200μg/mL,respectively.
