艾灸对佐剂性类风湿关节炎大鼠模型VEGF表达的影响

摘　　要：目的探讨艾灸对佐剂性类风湿关节炎大鼠血管内皮生长因子(VEGF)表达的影响。方法 Wistar大鼠60只,随机分为对照组、模型组与艾灸组,每组20只。模型组和艾灸组的大鼠于右足趾皮下注射福氏完全佐剂(CFA)0.15ml,艾灸组在造模第2天进行艾灸干预,第7、14、21、28天检测关节炎评分及左右足趾关节肿胀度;ELISA法检测血清中VEGF的含量,免疫荧光、Western blot方法检测踝关节滑膜VEGF的表达。结果模型组与对照组相比关节炎指数评分明显升高,而艾灸组比模型组明显降低(P<0.05);模型组与对照组相比,血清及踝关节滑膜中VEGF表达水平均显著升高,而艾灸组比模型组显著降低(P<0.05)。结论艾灸能够降低关节炎指数评分,其机制可能与下调VEGF表达有关。Objective To investigate the effect of moxibustion on the expression of vascular endothelial growth factor(VEGF) in adjuvant rheumatoid arthritis(RA) rats. Methods Sixty Wistar rats were randomly divided into control group, model group and moxibustion group, with 20 rats in each group. Rats in the model group and the moxibustion group were injected subcutaneously with 0.15 ml of Freund’s complete adjuvant(CFA) on the right toe. The moxibustion group was treated with moxibustion on the second day of modeling. On the 7 th, 14 th, 21 st and 28 th day, the arthritis score and the degree of swelling of the right and left joints were detected. The content of VEGF in serum was detected by ELISA, the expression of VEGF in the synovial membrane of the ankle was detected by immunofluorescence and Western blot. Results The arthritis index score was significantly higher in the model group than in control group, but significantly lower in moxibustion group than in model group(P<0.05);the expression level of VEGF in the serum and ankle synovium was significantly increased in the model group than in control group, but decreased in the moxibustion group than in model group(P<0.05). Conclusion Moxibustion can reduce arthritis index score in rats with adjuvant rheumatoid arthritis, which is related to down-regulation of VEGF expression.
