芍药 炙甘草不同配比对偏头痛模型大鼠的影响

摘　　要：目的观察芍药、炙甘草不同配伍比例对硝酸甘油致偏头痛模型大鼠血浆中一氧化氮(NO)、降钙素基因相关肽(CGRP)、五羟色胺(5-HT)含量的影响。方法将SD大鼠按体质量分成6组:正常对照组、模型对照组、盐酸氟桂利嗪(西比灵)组、芍药炙甘草1∶1组、芍药炙甘草2∶1组、芍药炙甘草3∶1组,除正常对照组、模型对照组灌胃0.9%氯化钠注射液外,其余组灌胃药物,7 d后,除正常组注射0.9%氯化钠注射液外,其余各组大鼠均皮下注射硝酸甘油复制偏头痛模型。观察大鼠行为学的表现,并检测大鼠血浆中NO、CGRP、5-HT的含量,评定不同配比芍药炙甘草治疗偏头痛的效果。结果除正常对照组之外,其余各组大鼠均出现了明显的耳红、频繁挠头等表现,芍药炙甘草3个配比均能推迟大鼠耳红出现时间,缩短耳红持续时间,减少挠头次数(P<0.05或P<0.01),芍药炙甘草3∶1组较其余比例组效果明显。与模型对照组相比,芍药炙甘草3个比例组大鼠血浆CGRP均有明显减少(P<0.01);芍药炙甘草3∶1组NO含量显著减少(P<0.01)、5-HT含量有显著升高(P<0.05)。结论芍药炙甘草不同配比对偏头痛治疗有效,配伍比例3∶1时效果最佳,本研究为芍药甘草汤更好地治疗偏头痛提供一定的实验依据。Objective To observe the effect of different dosage ratio of paeoniae glycyrrhiza on the content of nitric oxide(NO),calcitonin gene-related peptide(CGRP)and serotonin(5-HT)in plasma of migraine model rats induced by nitroglycerin.Methods SD rats were randomly divided into 6 groups:control group,model group,sibelium group,1∶1,2∶1 and 3∶1 groups of paeoniae&glycyrrhiza.After gavage by different medicine for 7 days,migraine rats were modelled by subcutaneous injection of nitroglycerin.The behavior of rats was observed and the contents of NO,CGRP and 5-HT in plasma were measured.Results Except for the control group,the other groups had the red ear and frequent scratching.The three dosage ratios of paeoniae and glycyrrhiza can delay the starting time of red ear,shorten the duration of red ear and reduce the number of scratching(P<0.05 or P<0.01).The effect of 3∶1 group was more obvious than that of the other ratio groups.The CGRP in plasma of the three ratio groups of paeoniae&glycyrrhiza was significantly reduced(P<0.01);the NO and 5-HT content in the 3∶1 group of paeoniae&glycyrrhiza weresignificantly reduced(P<0.01)and increased,respectively(P<0.05).Conclusion This study proved that the different dosage ratios of paeoniae&glycyrrhiza had the effectiveness on migraine treatment,and the 3∶1 of paeoniae&glycyrrhiza is the best ratio.It provided a certain experimental basis for the better treatment of migraine with paeoniae&glycyrrhiza decoction.
