摘  要:目的探讨加味芎蝎散对咳嗽变异性哮喘(CVA)大鼠咳嗽反应及气道平滑肌中IL8-CXCR1的干预作用。方法40只大鼠随机分为4组:正常组,CVA组,顺尔宁组,中药组。分别于2、4、6 d记录吸入辣椒素溶液后大鼠的咳嗽次数;采用酶联免疫法对气道灌洗液(BALF)Th1/Th2相关炎症分子表达进行观测;免疫组化法对气道壁血管指标CD31、气道平滑肌IL8、CXCR1、MMP9的表达影响进行研究。结果中药、顺尔宁组在不同程度上都能够减少CVA大鼠咳嗽次数,且中药组在起效时间及抑制程度上明显优于顺尔宁组;肺泡灌洗液中中药组及顺尔宁组TH1细胞分泌的炎症介质IFNγ升高,而TH2细胞分泌IL4、IL8降低;CVA组气道平滑肌组织MMP9和肺间质CD31及IL8、CXCR1表达增高,中药组及顺尔宁组表达较CVA组减低。结论加味芎蝎散在改善CVA大鼠咳嗽方面疗效确切;其作用机制可能与基于IL8-CXCR1靶点,从而调节TH1/TH2免疫失衡,抑制细胞迁移和血管新生有关。Objective To explore the effect of Jiawei Xiongxie Powder on expression of IL8-CXCR1 on airway smooth muscle and the cough response in CVA rats.Method Forty rats were randomly and equally divided into 4 groups:control group,CVA group,Montelukast Sodium group,Jiawei Xiongxie Powder group.The cough response caused by capsaicin was observed.The expression of Th1/Th2-related inflammatory molecules in bronchoalveolar lavage fluid(BALF)was observed by enzyme-linked immunosorbent assay(ELISA).Immunohistochemical method was used to study the expressions of CD31,IL8,CXCR1 and MMP9 in airway smooth muscle.Result Jiawei Xiongxie Powder group and Montelukast Sodium group can reduce the cough frequency of CVA rats in varying degrees,and the onset time and inhibition degree of Jiawei Xiongxie Powder group was significantly better than that of the Montelukast Sodium group.The expression of IFNγwas increased in Jiawei Xiongxie Powder group and Montelukast Sodium group,while IL4 and IL8 secreted by TH2 cells were decreased.The expressions of MMP9,CD31,IL8 and CXCR1 in airway smooth muscle tissue and pulmonary interstitial tissue of the CVA group were increased,while those of the Jiawei Xiongxie Powder group and Montelukast Sodium group were decreased.Conclusion Jiawei Xiongxie Powder has a definite effect on improving cough in CVA rats,and its mechanism may be related to regulating TH1/TH2 imbalance as well as inhibiting cell migration and angiogenesis based on IL8-CXCR1 target.
