摘  要:目的观察王氏连朴饮对急性肺损伤(acute lung injury,ALI)大鼠肺、肠组织TNF-αmRNA、GRmRNA表达,探讨该方对肠-肺轴保护作用的机制。方法将雄性Wistar大鼠36只(180 g)随机分成对照组、模型组、模型+地塞米松组、模型+王氏连朴饮大剂量组、模型+王氏连朴饮中剂量组、模型+王氏连朴饮小剂量组共计6组。PCR检测大鼠肺、肠组织中TNF-αmRNA、GRmRNA的水平。结果模型组大鼠肺、肠组织TNF-αmRNA表达明显升高(P<0.01),GRmRNA表达明显降低(P<0.01);中药组各剂量对TNF-αmRNA表达均有下调,其中大剂量组效果最显著(P<0.01)。对GRmRNA表达均有上调,其中大剂量组效果最显著(P<0.01)。与地塞米松组相比,王氏连朴饮大剂量组肺组织TNF-αmRNA、GRmRNA含量差异无统计学意义(P>0.05),中、小剂量组肺组织TNF-αmRNA含量差异显著(P<0.01)。大剂量组肠组织TNF-αmRNA、GRmRNA含量有差异(P<0.05),中、小剂量组肠组织TNF-αmRNA、GRmRNA含量差异显著(P<0.01)。结论王氏连朴饮对ALI大鼠肺、肠黏膜具有的保护作用可能源于对TNF-α和GR表达的调控作用。Objective To observe the expressions of TNF-αmRNA and GRmRNA in lung and intestine tissues of acute lung injury(ALI)rats before and after treatment with Wangshi Lianpu Drink and investigate the protective effect of this prescription on the intestinal-pulmonary axis.Methods Thirty-six male Wistar rats(180 g)were randomly divided into control group,model group,model+dexamethasone group,model+Wangshi Lianpu Drink high-dose group,model+Wangshi Lianpu Drink middle-dose group,model+Wangshi Lianpu Drink small-dose group,totally 6 groups.The levels of TNF-αmRNA and GRmRNA in rat lung and intestine tissues were detected by PCR.Results The expressions of TNF-αmRNA in the lung and intestine of the model group were significantly increased(P<0.01),and the expression of GRmRNA was significantly decreased(P<0.01).The TNF-αmRNA expression was down-regulated in each Wangshi Lianpu Drink dose group.The effect was significant(P<0.01).The expression of GR mRNA was up-regulated,with the highest dose group was mostly significant(P<0.01).Conclusion The protective effect of Wangshi Lianpu Drink on the intestinal-pulmonary axis of ALI rats may be due to the regulation of the expressions of TNF-αand GR.
