摘 要:目的基于内毒素特异质肝损伤模型,考察不同炮制程度的何首乌对大鼠肝脏的损伤作用,以优选何首乌最为安全的炮制工艺。方法采用无毒剂量的脂多糖(LPS,尾静脉注射2.8 mg/kg)制备内毒素特异质模型大鼠,模型大鼠和正常大鼠均灌胃给予12.96 g/kg不同炮制程度(0、8、16、24、32 h)的何首乌50%乙醇提取物,各给药1次,检测大鼠血浆中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)水平以及HE染色观察肝脏病理学改变。结果在灌胃给予不同炮制程度的何首乌50%乙醇提取物后,正常SD大鼠上,未见明显肝损伤作用,炮制24 h,32 h组ALT,AST值低于正常组;在内毒素特异质模型上,生首乌的ALT,AST值显著升高,炮制8 h,炮制16 h的ALT,AST值均高于正常组。结论何首乌以常压炮制32 h最为安全。Objective The objective of this study was to investigate the effects of Polygonum multiflorum radix on lipopolysaccharide(LPS)-mediated liver injury in rats following different degrees of Polygonum multiflorum radix processingin order to optimize Polygonum multiflorum radix processing approaches.Methods Rats received a non-fatal dose of LPS via caudal vein injection,after having been intragastrically administered 12.96 g/kg of 50%ethanol extract of Polygonum multiflorum radix that had been processed for a range of times(0,8,16,24,32 h).After 7 h,plasma samples and liver samples were collected for measurement of serum ALT/AST and histopathological assessment,respectively.Results Normal rats exhibited no significant liver injury.When Polygonum multiflorum radix samples had been processed for 24 h or 32 h.The mean ALT and AST values were lower relative to untreated controls.When rats also received LPS model,ALT and AST levels associated with Polygonum multiflorum radix were significantly increased,with those values of rats receiving Polygonum multiflorum radix prepared for 8 h and 16 h being higher than those in the normal control group.Conclusion The safest approach to Polygonum multiflorum radix processing is a 32 h processing approach conducted under normal pressure.
