摘  要:目的探讨大黄素对非酒精性脂肪性肝炎(non-alcoholic steatohepatitis,NASH)模型大鼠的治疗作用及对脂联素、脂联素受体的影响。方法雄性SD大鼠40只,随机分为空白组,模型组,大黄素高(40 mg·kg^-1)、低剂量(20 mg·kg^-1)组。空白组予普通饲料,其余各组予高脂饲料建模,时间共10周。造模后,处死动物,取材。观察各组大鼠体质量、肝湿重、肝指数,化学发光法检测大黄素对NASH模型大鼠谷丙转氨酶(glutamic pyruvic transaminase,ALT)、谷草转氨酶(millet straw transaminase,AST)、碱性磷酸酶(alkaline phosphatase,ALP)、谷氨酰转肽酶(glutamyl transpeptidase,GGT)、甘油三酯(triglycerides,TG)、胆固醇(cholesterol,TC)的影响,制作石蜡切片行苏木素-伊红染色(HE),油红O染色检测大黄素对NASH大鼠肝组织炎症损伤、脂肪变的影响,Western-blot检测各组大鼠肝脏内脂联素(adiponectin,ADPN)、脂联素受体2(adiponectin receptor-2,AdipoR2)蛋白表达,荧光定量PCR检测各组大鼠肝脏内ADPN,AdipoR2 mRNA表达。结果与模型组相比,大黄素能有效改善NASH模型大鼠肝脏脂质沉积,减轻炎症细胞浸润,减轻大鼠体质量,肝指数;大黄素高剂量组ALT、AST、ALP、GGT、TG、TC均有不同程度下降(P<0.05);相对模型组,大黄素高剂量组大鼠肝组织ADPN,AdipoR2蛋白,mRNA水平升高(P<0.05)。结论大黄素能通过上调NASH模型大鼠ADPN,AdipoR2表达,有效改善肝功能、血脂,改善NASH大鼠肝组织脂肪变程度,减轻肝脏炎症。Objective To investigate the effect of Rheum emodin on non-alcoholic steatohepatitis(NASH)model rats and its effects on adiponectin and adiponectin receptors.Methods Forty male SD rats were randomly divided into normal group,model group,high dose Rheum emodin(40 mg·kg^-1),low dose Rheum emodin(20 mg·kg^-1)group.The normal group was given normal feeding,and other groups were modeled for high-fat feed for a total of 10 weeks.After the model was completed,the animals were sacrificed and the materials were taken out.The body weight,liver wet weight and liver index were observed in each group.ALT,AST,ALP,GGT,TG and TC were tested by Chemiluminescence.HE and Oil red staining were used to detect the effects of Rheum emodin on hepatic inflammatory and fatty changes in model NASH rats.Western-blot was used to detect the protein of adiponectin and adiponectin receptor-2 in livers of rats in each group.The expressions of mRNA of ADPN and AdipoR2 in the liver of each group were detected by QPCR.Results Compared with the model group,Rheum emodin can effectively improve the liver lipid deposition in NASH rats and reduce the infiltration of inflammatory cells.ALT,AST,ALP,GGT,TG and TC levels in Rheum emodin high-dose group decreased to varying degrees(P<0.05).Compared with the model group,the emodin high-dose group’s liver ADPN,AdipoR2 protein and mRNA levels were significantly increased(P<0.05).Conclusion Rheum emodin can up-regulate the expressions of ADPN and AdipoR2 in NASH model rats,effectively improve liver function and blood lipids,improve the degree of hepatic steatosis in NASH rats and reduce liver inflammation.
