摘 要:目的建立同时测定黄芩饮片中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素含量的方法,对不同产地规格临床中药饮片黄芩进行质量评价。方法运用高效液相色谱法,采用Thermo Acclaim120-C18(4. 6 mm×250 mm,5μm)色谱柱,以乙腈(A)、0. 2%磷酸溶液(B)为流动相梯度洗脱,流速为1. 0 m L/min,进样量为10μL,检测波长为280 nm,柱温为30℃。结果黄芩苷、汉黄芩苷、黄芩素、汉黄芩素的质量分别在0. 1500~1. 1998μg(r=0. 9994)、0. 0499~0. 3994μg(r=1. 0000)、0. 0025~0. 0202μg(r=0. 9989)、0. 0025~0. 0198μg(r=1. 0000)范围内具有良好的线性关系。20批样品中上述4种黄酮类成分的平均含量范围分别为60. 4~148. 58 mg/g、14. 77~30. 99 mg/g、0. 99~26. 82 mg/g、0. 74~6. 18mg/g。结论临床中药饮片黄芩质量状况差异很大,本实验建立并优化的黄芩饮片定量分析方法,可为不同规格黄芩饮片的质量评价及临床用药提供依据。Objective:To establish an HPLC method for simultaneous determination of baicalin,wogonoside,baicalein and wogonin in Scutellaria baicalensis slices and evaluate the quality of Scutellaria baicalensis slices with different origins and specifications.Methods:Using high performance liquid chromatography(HPLC),Thermo Acclaim120-C18(4.6 mm×250 mm,5μm)column was used for the gradient elution of acetonitrile(A)and 0.2%phosphoric acid solution(B).The flow rate was 1 mL/min,the sample volume was 10μL,the detection wavelength was 280 nm,and the column temperature was 30℃.Results:The quality of baicalin,wogonoside,baicalein and wogonin had good linear relationship in the range of 0.1500-1.1998μg(r=0.9994),0.0499-0.3994μg(r=1.0000),0.0025-0.0202μg(r=0.9989),0.0025-0.0198μg(r=1.0000).The average contents of the above four flavonoids in the 20 batch samples ranged from 60.4 mg/g to 148.58 mg/g,14.77 mg/g to 30.99 mg/g,0.99 mg/g to 26.82 mg/g,0.74 mg/g to 6.18 mg/g,respectively.Conclusion:The quality of Radix Scutellariae in clinical Chinese medicine varies greatly and the experiment established quantitative analysis method of Scutellaria baicalensis pieces and provided the basis for quality evaluation and clinical medication of Scutellaria baicalensis slices with different specifications.
