摘 要:目的研究橘叶提取物对实验性自身免疫性睾丸炎(EAO)模型小鼠睾丸组织形态及血清白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量的影响。方法采用单侧睾丸注射冰醋酸法建立小鼠EAO模型,随机分为空白组、模型组、橘叶乙醇提取物组、环己烷组、乙酸乙酯组、正丁醇组和水部位组。光镜下观察睾丸组织结构变化,ELISA法检测血清IL-1β和TNF-α浓度。结果给药3周后,橘叶乙醇提取物、正丁醇部位和水部位组能明显改善睾丸组织病变,减轻炎症反应。橘叶给药组除环己烷部位外,其余各组均能明显降低EAO小鼠血清中炎症因子IL-1β水平,与模型组比较,差异有统计学意义(P <0. 01)。此外,乙醇提取物组、正丁醇部位组中TNF-α含量也明显降低(P <0. 05,P <0. 01)。结论橘叶乙醇提取物、乙酸乙酯、正丁醇及水部位对小鼠EAO的发生均具有拮抗作用,其中,正丁醇部位抗EAO活性较明显,这可能与抑制炎性因子IL-1β和TNF-α释放有关。Objective:To study the effects of the extracts from leaves of Citrus reticulata on the condition of the testicle histomorphology and the levels of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)of experimental autoimmune orchitis(EAO)in mice.Methods:The EAO model in mice was established by the method of unilateral testicular injection with glacial acetic acid,and then the mice were randomly divided into normal saline group,model group,and the groups of EtOH extracts,cyclohexane part,EtOAc soluble fraction,n-BuOH-soluble part and water layer from leaves of Citrus reticulata.Moreover,the testicular histology was observed under optical microscope,and the levels of IL-1βand TNF-αwere evaluated by ELISA.Results:Pathological changes were apparently improved and the inflammatory response had a weak tendency as treated by EtOH extracts,n-BuOH layer and water-soluble part from leaves of Citrus reticulata 3 weeks after operation.Moreover,the levels of IL-1βwere decreased obviously in the experimental groups from the leaves of Citrus reticulata except for the cyclohexane part.Compared with the model group,it had significant difference(P<0.01).Besides,the levels of TNF-αwere decreased obviously as treated by EtOH extracts and n-BuOH layer(P<0.05,P<0.01).Conclusion:The groups of EtOH extracts,EtOAc soluble fraction,n-BuOH-soluble part and water layer from leaves of Citrus reticulata have anti-EAO activities in mice,and the n-BuOH-soluble part has stronger effect,which may be related to the inhibition of the release of IL-1βand TNF-α.
