摘  要:目的:通过观察8周大强度间歇运动(HIIT)和有氧运动对肥胖大鼠骨骼肌5’单磷酸腺苷激活蛋白激酶(AMPK)和过氧化物酶体增殖受体γ辅助激活因子α(PGC-1α)表达的影响,探讨不同运动时AMPK/PGC-1α通路在减脂过程中的骨骼肌适应机制。方法:高脂饮食诱导的SD肥胖大鼠建模成功后选取30只,随机分为安静对照组(OC组)、有氧运动组(OA组)和大强度间歇运动组(HIIT组),每组10只。OC组大鼠不运动;OA组和HIIT组均进行8周0坡度跑台运动,5次/周,高脂饲料自由饮食。运动前测试VO2max并确定相应跑速,4周后调整一次跑速。OA组以60%~70%VO2max相应速度运动60 min;HIIT组先以70%VO2max相应速度热身,再以90%、50%VO2max相应速度交替运动,最后以70%VO2max相应速度恢复,运动时间以与OA组运动距离一致计算。训练期间每周固定时间测量体重。运动8周后恢复24 h腹主动脉取血,测试血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C);取肾周和附睾脂肪称重,计算脂体比([肾周脂肪重量+附睾脂肪重量)/体重*100%];采用实时荧光定量PCR及Western Blot测试腓肠肌AMPK、PGC-1α基因和蛋白表达。结果:(1)与OC组相比,HIIT组和OA组大鼠体重、肾周和附睾脂肪重量均显著降低(P<0.01或P<0.05);与OA组相比,HIIT组肾周和附睾脂肪重量分别降低21.3%和22.4%;HIIT组脂体比较OC组显著下降,较OA组下降22.2%。(2)与OC组相比,HIIT组TC、TG和LDLC均下降(P<0.01或P<0.05);与OA组相比,HIIT组TC、TG和LDLC分别降低13.9%、21.4%和17.4%。(3)HIIT组AMPK mRNA表达较OC组上调9.6%(P<0.05),较OA组上调104%(P<0.01);与OC组相比,OA组AMPK mRNA表达量下降(P<0.01)。HIIT组和OA组AMPK蛋白表达较OC组上调11%和10%(P<0.05)。HIIT组和OA组PGC-1αmRNA表达较OC组分别上调520%和860%(P<0.01);HIIT组较OA组下调35.4%(P<0.01)。HIIT组和OA组PGC-1α蛋白表达量较OC组分别上调9%和18%。结论:(1)Objective To investigate the effect of high-intensity interval training(HIIT)and aerobic exercise on skeletal muscle adaption during weight loss by activating AMPK/PGC-1αsignaling pathway in obese rats.Methods Totally 30 diet-induced obese Sprague-Dawley rats were randomly divided into a control group(OC),an aerobic exercise group(OA)and an HIIT group(HIIT),each of 10.The rats in OA and HIIT groups underwent 8-week aerobic exercise and HIIT respectively 5 days per week.The VO2max of rats were measured before training to determine the running speed.In each training session,the OA group ran at a speed corresponding to 60%-70%VO2max for 60 minutes;the HIIT group ran at 90%VO2max interspersed with recovery periods at 50%VO2max,repeated 6 times.The running distance was matched between OA and HIIT group.During the training,the weight was mea sured every week.The perirenal and epididymal fat mass were taken 24 hours after the 8-week inter vention.The serum total cholesterol(TC),triglyceride(TG),high density liptein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)were detected using the enzymatic method,and the gene and protein expressions of adenosine 5’-monophosphate-activated protein kinase(AMPK)and per oxisome proliferators-activated receptorγco-activator 1 alpha(PGC-1α)were determined using the re al-time fluorescence quantitative polymerase chain reaction(RT-PCR)and Western blotting respective ly.The perirenal and epididymal fat were measured and the weight of perirenal and epididymal fat against the body weight was calculated.Results The average weight of perirenal and epididymal fat of HIIT and OA groups were significantly lower than that of OC group(P<0.01,P<0.05).The weight of perirenal and epididymal fat and the body weight ratio(fat weight/body weight)of the HIIT group de creased by 21.3%,22.4%and 22.2%compared with OA group.The average TC,TG and LDL-C lev els of HIIT group were significantly lower than OC group(P<0.01,P<0.05).The average TC,TG and LDL-C levels of HIIT group decreased by
