青海大通北川河源区鱼类舌状绦虫和双线绦虫的分子鉴定及系统发育关系研究  被引量:3

摘  要:通过PCR方法扩增青海大通北川河源区国家级自然保护区河流中鱼体内寄生的舌状绦虫和双线绦虫CoxI、ItsI和NadI基因部分序列,并进行分子鉴定,分析3种基因的同源性,利用这3种基因进行分子进化和系统发育关系研究。结果显示,经过分子鉴定,寄生在鱼类肠道中的绦虫为舌状绦虫,寄生在鱼类腹腔中的绦虫为双线绦虫;舌状绦虫裂头蚴克隆的CoxI、ItsI和NadI基因序列分别与EU241312(法国)、KC900974(伊朗)和AF524046(中国)的核苷酸序列同源性为97.8%、99.3%和84.2%,双线绦虫克隆的CoxI、ItsI和NadI基因序列分别与EU241309(法国)、KC900994(伊朗)和AF254123(中国)的核苷酸序列同源性为89.6%、98.9%和99.5%;舌状绦虫与双线绦虫3种基因序列比对发现,CoxI基因序列同源性为83.2%,ItsI基因序列同源性为91.1%,NadI同源性为71.8%。同时,利用GenBank中其他种属绦虫的CoxI、ItsI和NadI基因序列构建系统发育树,均与已鉴定的舌状绦虫和双线绦虫聚在同一支上,与其他绦虫所属分支相距较远,从构建的系统发育树可以看出舌状绦虫和双线绦虫分属两个不同种类,且与舌状绦虫中国广州分离株种属亲缘关系较近。初步阐明了鉴定的舌状绦虫和双线绦虫分离株与其他种属之间的系统发育关系。We used PCR to amplify the partial sequence of CoxI, ITSI and NadI genes of Ligula intestinalis and Digramma interuota parasitizing in the abdominal cavity of fish in the rivers of the national nature reserve of Qinghai Datongbeiheyuanqu Nature Reserve molecular identification to analyze homology of the three genes and the molecular evolution and phylogenetic relationships of the three genes. The results showed that the tapeworm, which is parasitic in the intestines of fish, was identified as ligula intestinalis, and the tapeworm in the fish’s abdominal cavity was identified as Digramma interuota.For ligula intestinalis pleroceroid its CoxI、ItsI and NadI gene Nucleotide sequence homology was 97.8%, 99.3% and 84.2% with EU241312(France), KC900974(Iran) and AF524046(China), Digramma interuota that its CoxI、ItsI and NadI gene Nucleotide sequence homology was 89.6%、98.9% and 99.5% with EU241309(France), KC900994(Iran) and AF254123(China)respectively. Comparison of the 3 gene sequences of Ligula intestinalis and Digramma interuota showed that CoxI、ItsI and NadI gene sequence homology was 83.2%,91.1% and 71.8% respectively. The phylogenetic relationship between the identified Ligula intestinalis and digramma interuota with other tapeworm species was elucidated.
