滇金石斛中4个对映海松烷二萜化合物的分离鉴定及其抗龋作用研究

摘　　要：目的研究滇金石斛Flickingeria albopurpurea Seidenf中的化学成分,对分离得到的对映海松烷二萜类物质进行致龋相关细菌的抑制作用研究。方法综合运用硅胶、Sphedex LH-20凝胶柱色谱及半制备Rp-HPLC对化学成分进行分离,运用多种现代波谱技术进行结构鉴定;利用MBECTM-Device考察分离到的化合物对变形链球菌、血链球菌、唾液链球菌、内氏放线菌、黏性放线菌、乳杆菌等6种致龋菌生长的影响,并测定化合物对细菌的最小抑菌浓度(MIC)和细菌生物膜的最低清除浓度(MBEC)。结果从滇金石斛中分离鉴定出4个对映海松烷二萜化合物,它们对6种致龋菌均有不同程度的抑制作用,其中对变形链球菌抑制作用最强。结论4个化合物均为首次从滇金石斛中分离得到,均具有抑制致龋菌的活性,是潜在的抗龋活性天然产物。Objective To study the chemical constituents of Flickingeria albopurpurea Seidenf and their effects on the growth of the main cariogenic bacteria.Methods The silica gel column chromatography,Sephadex LH-20 column chromatography and semi preparative RP-HPLC were used to isolate and purify the compounds from the stems of Flickingeria albopurpurea,and their structure was elucidated by spectroscopic data analysis.MBECTM-Device was used to study the effects on the growth of Streptococcus,Streptococcus sanguis,Streptococcus salivarius,actinomycete Neisseria,actinomycete viscosus and Lactobacillus.The minimal inhibitory concentration(MIC)and the minimal biofilm eradication concentration(MBEC)of the test compounds were determined.Results Four ent-pimarane diterpenoids were isolated and identified.All compounds showed inhibitory activities on the growth of cariogenic bacteria and the forming of biofilm,which had the strongest inhibition on Streptococcus mutans.Conclusion Compound 1-4 were isolated from Flickingeria albopurpurea for the first time,which may be developed into anti-caries agent for their inhibition on the cariogenic bacteria.
