摘  要:目的改进固氮菌固氮能力和解磷菌解磷能力的测定方法。方法通过重结晶K 2S 2O 8使得采用《水质总氮的测定碱性过硫酸钾消解紫外分光光度法》(HJ 636-2012)测定空白样品的校正吸光度值达到标准(吸光度<0.03);通过调整显色酸度使得按照钼锑抗分光光度法测定解磷菌发酵液中有效磷浓度时显色完全。结果K 2S 2O 8重结晶4次,空白样品吸光度值可达到要求。按照钼锑抗分光光度法测定解磷能力时,加入解磷菌发酵液后,调整酸度至4.4,再定容显色,可使磷钼蓝显色完全。结论经完善后的固氮、解磷能力的测定方法达到标准中的要求,测定结果更准确可靠。Objective To improve the determination methods of nitrogen fixation and phosphorus dephosphorization ability of nitrogen-fixing bacteria and phosphorus dephosphorization bacteria.Methods The blank value was standardized(A<0.03)through recrystallizing potassium persulfate by adopting the method of total nitrogen detection in water using alkaline potassium persulfate digestion ultraviolet spectrophotometry(HJ 636-2012).The molybdenum anti-spectrophotometric method was completely developed by adjusting the color development acidity.Results The potassium persulfate was recrystallized four times,the blank value could meet requirement.In the determination of the ability of phosphate solubilization by the antimony molybdenum spectrophotometric method,after adding the phosphate-dissolving bacteria fermentation broth,adjusting the acidity to 4.4,and then developing the color at a constant volume,phosphomolybdenum blue would be completely colored.Conclusion The improved method for determination of nitrogen fixation and phosphate-dissoving ability is more accurate and reliable.
