摘 要:目的探讨红景天苷对急性一氧化碳中毒心肌损伤的保护作用。方法取SD封闭大鼠72只,随机分为A、B、C 3组,分别给予不同浓度的一氧化碳染毒,制作一氧化碳中毒模型,A组为轻度中毒组,B组为中度中毒组,C组为重度中毒组。A、B、C组内再随机分为A1、A2,B1、B2,C1、C2亚组。A1、B1、C1亚组均给予红景天苷(10mg/kg)尾静脉注射,A2、B2、C2亚组均给予等量的0.9%氯化钠溶液尾静脉注射。记录24h生存率、心律失常发生率,检测血天冬氨酸转氨酶(AST)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)和肌钙蛋白I(cTn Ⅰ)。染毒后24h取心脏组织测量心脏湿重,并行电镜检查。结果 C1亚组与C2亚组比较,AST、LDH、CK、CK-MB、cTn Ⅰ均显著降低(均P<0.05),生存率升高、心律失常发生率降低、心脏湿重降低,差异均有统计学意义(均P<0.05)。B2亚组与B1亚组比较,AST、LDH、CK、CK-MB均明显升高,差异均有统计学意义(均P<0.05)。A1与A2亚组之间上述指标无明显差异(均P>0.05)。心脏组织电镜检查显示B1、C1亚组较B2、C2亚组心肌组织改变明显减轻。结论红景天苷对急性一氧化碳中毒导致的心肌损伤有保护作用。Objective To explore the effect of Salidroside on myocardial damage induced by acute carbon monoxide poisoning(ACMP). Methods 72 sealant-grade Sprague-Dawley rats were randomly divided into group A, B and C.Mild, moderate and serious ACMP models were established in group A, B and C using different concentrations of carbon monoxide. Each group was divided into two subgroups as group A1 and A2,B1 and B2, C1 and C2. Salidroside(10 mg/kg) was injected into tail vein in group A1,B1 and C1. Equivalent normal saline was injected in group A2, B2 and C2. The survival rate and arrhythmias of 24 hours were recorded. Serum AST, LDH, CK, CK-MB and cTn Ⅰ were measured,and rat hearts were taken for measurement of wet weight and electron microscope examination at 24 h after poisoning of carbon monoxide. Results AST,LDH,CK,CK-MB and cTn I as well as arrhythmias and heart wet weight were significantly lower, and the survival rate was significantly higher in group C1 than group C2(all P<0.05). AST, LDH, CK and CK-MB were significantly lower in group B1 than group B2(all P<0.05). There was no significant difference of above parameters between group A1 and A2(all P >0.05). The electron microscopic findings showed the changes of myocardial tissues were relatively milder in group C1 and B1 than in group C2 and B2, respectively. Conclusion Salidroside may improve myocardial damage induced by acute carbon monoxide poisoning.
