摘 要:基于16S rRNA基因测序技术对24份东北马鹿冬季粪便中细菌的结构和多样性进行分析,试图揭示东北马鹿冬季肠道微生态,为该物种保护提供新的见解。结果表明东北马鹿粪便样品中菌群分属于21门33纲74目131科306属,在门水平上,马鹿肠道菌群主要由厚壁菌门(Firmicutes)和拟杆菌门(Bacteroidete)构成,相对丰度分别为79.43%和6.64%;在属水平上,马鹿肠道菌群主要由Ruminococcaceae_UCG-005和Christenseenellaceae_R-7_group构成,相对丰度分别为21.51%和7.56%。以97%的序列一致性为划分标准进行聚类得到2220个OTUs,雄性样本中共有1873个OTUs,雌性样本中共有2143个OTUs,其中77个OTUs为雄性样本所特有,347个为雌性样本所特有。对比不同性别东北马鹿肠道菌群,通过LEfSe多级物种差异判别分析,丰度具有显著性差异的菌类包括:Sphingomonadales目、Kaistiaceae科、Sphingomonadaceae科、Muribaculaceae科、Xanthobacteraceae科、Defluviitaleaceae科、Kaistia属、Papillibacter属、Sphingomonas属、Alloprevotella属、GWE 2_31_10、Defluviitaleaceae_UCG_011、Eubacterium_ruminantium_group。We used high-throughput sequencing of the 16S rRNA gene to analyze the structure and diversity of the microbiota of 24 fecal samples collected from red deer(Cervus elaphus xanthopygus)during winter and in an effort to describe the intestinal microbiota of red deer.The gut microbiota community comprised 21 phyla,33 classes,74 orders,131 families and 306 genera.At phylum level,Firmicutes(79.4%)was most abundant,followed by Bacteroidetes(6.6%).At genus level,Ruminococcaceae_UCG-005(21.5%)was most abundant,followed by Christenseenellaceae_R-7_group(7.6%).Based on a distance matrix of 97%of similarity,2220 operational taxonomic units(OTUs)were identified,1873 OTUs were identified in male samples and 2143 OTUs were identified in female samples,77 of 1873 OTUs were unique to male samples and 347 of 2143 OTUs were unique to female samples.The results showed that the alpha diversity and beta diversity between sexes were not significantly different.At family and genus level,significant differences were detected between sexes,including Kaistiaceae,Sphingomonadaceae,Muribaculaceae,Xanthobacteraceae,Defluviitaleaceae,Kaistia,Papillibacter,Sphingomonas,Alloprevotella,GWE 2_31_10,Defluviitaleaceae_UCG_011 and Eubacterium__ruminantium_group.
