摘  要:目的:探讨复方中药金水清合剂对IgA肾病大鼠TGF-β1、CTGF、VEGF蛋白及mRNA表达的影响。方法:4周龄SPF级SD大鼠40只,随机分为正常组、模型组、治疗1组、治疗2组、治疗3组,复制大鼠IgA肾病模型,观察组在模型组的基础上,分别于第5周、第7周、第9周,给予金水清合剂灌胃治疗,1次/d,至第12周末,HE及PAS染色观察肾脏病理学改变,Western-blot检测肾组织TGF-β1、CTGF、VEGF蛋白的表达,RT-PCR检测TGF-β1、VEGF及CTGF mRNA的表达。结果:正常组大鼠肾组织结构完整清晰,模型组大鼠可见肾小球系膜区增宽,系膜基质、系膜细胞中到重度增生,观察组病变程度比模型组轻,治疗1组改善最为明显。与正常组比较,模型组TGF-β1、CTGF、VEGF蛋白的表达升高,且差异有统计学意义(P<0.05);与模型组比较,观察组表达降低,且差异有统计学意义(P<0.05);观察组间比较差异有统计学意义(P<0.05);与正常组比较,模型组TGF-β1、CTGF、VEGF mRNA的表达升高,且差异有统计学意义(P<0.05);与模型组比较,观察组表达降低,且差异有统计学意义(P<0.05);观察组间比较,差异有统计学意义(P<0.05)。结论:金水清合剂能显著改善IgA肾病大鼠肾脏损害,早期治疗改善最为明显,其作用机制可能为下调TGF-β1、CTGF、VEGF蛋白及mRNA的表达。Objective:To investigate the effects of Jinshuiqing Mixture on the expression of TGF-β1,CTGF,VEGF protein and mRNA in IgA nephropathy rats.Methods:A total of 404-week-old SPF SD rats were randomly divided into a normal groups,a model group,treatment group 1,treatment group 2 and treatment group 3.Rat IgA nephropathy model was copied.Based on the model group,the treatment group was given Jinshuiqing Mixture by gavage once per day in the 5th week,the 7th week and the 9th week respectively till the end of 12th week.Pathological changes in kidney were observed by HE staining and PAS staining.The protein expression of TGF-β1,CTGF and VEGF was detected by Western blot,and the expression of TGF-β1,VEGF and CTGF and mRNA was detected by RT-PCR.Results:In the normal group,the renal tissue structure was complete and clear.In the model group,mesangial area was widened,and mesangial cells and mesangial matrix were moderately to severely proliferated.The degree of lesions in the treatment group was lighter than that in the model group.And the improvement of treatment group 1 was the most obvious one.Compared with the normal group,the expression of TGF-β1,CTGF and VEGF protein in the model group increased significantly and the difference was statistically significant(P<0.05);compared with the model group,the expression in the treatment group decreased greatly and the difference was statistically significant(P<0.05);compared among the treatment groups,and the difference was statistically significant(P<0.05);compared with the normal group,the expression of TGF-β1,CTGF,VEGF mRNA in the model group was significantly higher,and there was significant difference(P<0.05);compared with the model group,the expression in the treatment group was lower,and the difference was statistically significant(P<0.05);compared among the treatment groups,and the difference was statistically significant(P<0.05).Conclusion:Jinshuiqing Mixture can significantly improve the renal damage of IgA nephropathy rats,and early treatment improvement is most
