摘 要:自1986年第一个抗体药物OKT-3上市以来,越来越多的治疗性单克隆抗体进入临床研究并获批上市,利用治疗性单克隆抗体进行靶向治疗已经成为对抗癌症、病毒感染以及免疫性疾病的有效手段。随着细胞工程技术和基因工程技术的进步,为了更好的发挥单克隆抗体的治疗效果,研究人员已经进行了大量的研究来探索抗体的结构性质与其功能之间的联系,其中一个关键领域就是通过Fc工程改造来调节抗体与免疫系统的相互作用。本文将针对IgG结构、IgG抗体Fc区域的作用机理、IgG抗体亚型及Fc区功能差异、Fc区域的改造策略及Fc改造抗体的临床应用及发展前景进行综述。Since the launch of the first antibody drug OKT-3 in 1986,more and more therapeutic monoclonal antibodies have been enrolled in clinical research and approved for marketing.Targeted treatment with therapeutic monoclonal antibodies has become an effective means for the therapy of cancer,viral infection and immune disease.With the progresst of cell engineering technology and genetic engineering technology,researchers have conducted a lot of studies to explore the relationship between the structural of antibodies and their functions,and now more and more modification has been carried out to make therapeutic antibodies more effectively.One of the key researches is to regulate the interaction between antibodies and the immune system through Fc modification engineering.This paper reviews the IgG structure,the mechanism of action of Fc region in IgG antibody,IgG antibody subtypes and their functional differences in Fc region,the modification strategies of Fc region and the clinical application and development prospect of Fc modified antibodies.
