摘 要:目的:观察黄芪葛根汤总黄酮与总皂苷及其配伍对糖尿病大鼠肝脏糖代谢的影响。方法:将86只SD大鼠随机分为正常组、模型组、阳性对照组、总黄酮组、总皂苷组、总黄酮与总皂苷组。采用一次性腹腔注射STZ(按46 mg·kg-1)诱导糖尿病大鼠模型。血糖仪检测血糖水平,RTPCR法检测肝组织IRS-2、PI3K、Akt2、GLUT-4、Gsk-3βmRNA表达。结果:与正常组比较,模型组大鼠血糖明显升高,肝组织IRS-2、PI3K、Akt2、GLUT-4 mRNA水平降低(P<0.05);Gsk-3βmRNA水平显著升高(P<0.05)。总黄酮、总皂苷均有降糖作用,但无交互作用(P=0.064>0.05)。总皂苷能有效提升IRS-2 mRNA水平,总黄酮提升GLUT-4且降低Gsk-3βmRNA水平显著(P<0.05),两药存在交互作用,但合用不如单一用药;总黄酮、总皂苷均能提升PI3K、Akt2 mRNA水平,对PI3K mRNA表达则无交互作用。结论:总皂苷的降糖机制可能与IRS-2 mRNA表达有关,总黄酮与Gsk-3β、GLUT-4 mRNA表达有关,两组分相伍之降糖作用呈现各自为用的现象。Objective:To observe the effect of total flavonoids and total saponins of Huangqi Gegen Decoction and its compatibility on liver glucose metabolism in diabetic rats.Methods:86 SD rats were randomly divided into normal group,model group,positive control group,total flavonoid group,total saponin group,total flavonoid and total saponin group.A single intraperitoneal injection of STZ(at 46 mg·kg-1) was used to induce a diabetic rat model.The blood glucose level was detected by a blood glucose meter,and the expression of IRS-2,PI3 K,Akt2,GLUT-4,and Gsk-3β mRNA in liver tissues was detected by RTPCR.Results:Compared with the normal group,the blood glucose of the model group was significantly increased,and the levels of IRS-2,PI3 K,Akt2,and GLUT-4 mRNA in liver tissues were decreased(P<0.05);Gsk-3β mRNA levels were significantly increased(P<0.05).Total flavonoids and total saponins all had hypoglycemic effects,but no interaction(P=0.064>0.05).Total saponins can effectively increase the level of IRS-2 mRNA,total flavonoids significantly increase GLUT-4 and reduce Gsk-3β mRNA levels(P<0.05).There is an interaction between the two drugs,but the combination is not as good as a single drug;total flavonoids and total saponins can increase PI3 K,Akt2 mRNA level,but no interaction on PI3 K mRNA expression.CONCLUSION:The hypoglycemic mechanism of total saponin may be related to the expression of IRS-2 mRNA,and the total flavonoids are related to the expression of Gsk-3β and GLUT-4 mRNA.The two components have a hypoglycemic effect.
